摘要 [目的]应用DNA条形码技术对宁波口岸入境的24种有瓣蝇类进行分类研究。[方法]对有瓣蝇类样品的CO I基因片段进行了序列测定、比对和分析,使用MEGA6.0软件进行系统发育分析。[结果]24种有瓣蝇类CO I基因片段系统发育情况在科、属的水平上基本与形态学分类相同,在种的水平上不能够完全区分近缘种。[结论]单独用DNA条形码技术进行有瓣蝇类分类鉴定是不妥当的,需与形态学分类方法相密合,可作为形态分类方法的有益补充。 [Objective] A study on the classification of 24 species of Calyptratae entering Ningbo port using DNA barcoding technique was carried out.[Method] The CO I genes of the 24 species of Calyptratae were first sequenced.Based on the comparison and analysis of the obtained sequences,the phylogenetic tree was constructed using MEGA6.0.[Result] The cluster analysis showed the classification of the 24 species of Calyptratae was consistent with the morphological classification at the family and genus levels.However,the cluster analysis could not fully distinguish the closely-related species.[Discussion] The DNA barcoding technique cannot be singly used for classifying and identifying Calyptratae.It should be combined with morphological classification methods,and can be treated as a beneficial supplement for morphological classification methods.
