目的研究4种PM2.5中的矿物粉尘对V79细胞的毒性作用。方法将不同浓度的4种矿物粉尘分别作用V79细胞24h后,用噻唑蓝(MTT)试验测定细胞的存活率,检测上清液中白细胞介素-6(IL-6)含量,彗星试验(SCGE)检测细胞DNA的损伤程度。结果 4种矿物粉尘均对V79细胞存活率产生影响,随矿物浓度的增高,细胞存活率逐渐降低;4种矿物粉尘均能使V79细胞培养液上清液中的IL-6含量升高。4种矿物粉尘对V79细胞的DNA损伤呈现剂量—效应关系,矿物粉尘浓度越高,DNA损伤越明显。结论 4种PM2.5中的矿物粉尘对V79细胞均能产生细胞毒性,且能够引起V79细胞IL-6分泌的增加,对细胞DNA产生明显的损伤效应,PM2.5中的矿物粉尘可能通过炎症因子的释放和DNA的损伤等机制对细胞产生毒性作用。

Objective To study the toxic effects of PM2.5 mineral dusts to V79 cells,in order to explore its action mechanism.Methods The V79 cells was exposed to four kinds of mineral dusts with different concentrations for about 24 hours.The survival rate of V79 cells was measured by MTT assay,the supernatant fluid interleukin-6（il-6）levels were tested,and the DNA damage of V79 cells was detected by the SCGE.Results Compared to the control group,all the four kinds of the minerals dusts had impacts on the survival rate of the V79 cells.With the increasing concentration of the mineral dusts,the survival rate of V79 cells decreases gradually.The four kinds of the mineral dusts could rise the interleukin（il-6）levels of V79 cells culture supernatant.The four kinds of the minerals dusts could damage DNA of the V79 cells and the doseresponse relationship was found.The higher concentration of the mineral dusts,the more obvious of the DNA damage.Conclusions The four main composition of the PM2.5 mineral dusts can produce toxic effects on V79 cells,increase the secretion of IL-6,have damage to DNA.Then it comes to the conclusion that PM2.5mineral dusts may have toxic effect on cells through some mechanisms such as the release of inflammatory factors and DNA damage.
