白背叶根含药血清对脂多糖诱导下RAW264.7细胞分泌的影响 被引量:4

摘要 目的:观察不同浓度的白背叶根含药血清对脂多糖(lipopolysac-charide,LPS)诱导下RAW264.7细胞分泌的影响。方法:制作高、中、低浓度白背叶根含药血清,以CCK-8法检测不同浓度白背叶根含药血清对RAW264.7细胞活力的影响。以脂多糖刺激生长良好的RAW264.7细胞,建立细胞炎症模型,检测白背叶根含药血清对细胞上清液中炎症介质一氧化氮(NO)、前列腺素E2(PGE2)、白三烯B4(LTB4)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)含量的影响。结果:不同浓度的白背叶根含药血清均对RAW264.7细胞活力无影响;与模型组比较,10%、5%的白背叶根含药血清可显著抑制NO的释放(P<0.05或P<0.01),10%白背叶根含药血清可显著抑制LTB4的分泌(P<0.05),2.5%白背叶根含药血清可显著抑制IL-1β的分泌(P<0.05);但不同浓度的白背叶根含药血清对PGE2、TNF-α的分泌无显著影响。结论:白背叶根的抗炎机制可能是通过抑制炎症反应中NO,LTB4,IL-1β的释放而实现。 Objective: To observe effect of medicated serum containing the root of Mallotus apelta(Lour.) Muell. Arg. at different concentration on secretion activity in LPS-stimulated RAW264.7 cell. Methods: The medicated serum containing the root of Mallotus apelta (Lour.) Muell. Arg. of high, medium and low concentrations were prepared in order to detect the effect of it on RAW264.7 cell viability by CCK-8 method. LPS was used to stimulate the well- grown RAW264.7 cells so as to establish cell inflammation model. The effect of medicated serum containing the root of Mallotus apelta (Lour.) Muell. Arg. on content of inflammatory mediator [Nitric oxide (NO) ,prostaglandin E2 (PGE2), leukotriene B4 (LTB4), interleukin - 1β (IL- 1β), tumor necrosis factor -α (TNF-α) ] in cell supernatant was detected in comparison with model group. Results: The medicated serum containing the root of Mallotus apelta (Lout.) Muell. Arg. at different concentration had no effect on cell viability. Compared with model group, 10% and 5% medicated serum containing the root of Mallotus apelta (Lour.) Muell. Arg could inhibit the release of NO obvi- ously (P〈0.05 or P〈0.01 );2.5% medicated serum containing the root of Mallotuz apelta (Lour.) Muell. Arg. could inhibit the release the secretion of IL-1β obviously (P〈0.05). However,the medicated serum containing the root of Mallotus apelta(Lour. ) Muell. Arg. at different concentration had no obvious effect on the secretion of PGE2,TNF-α. Conclusion: The anti-inflammatory mechanism of the root of Mallotus apelta (Lour.) Muell. Arg. is likely to be achieved by inhibiting the release of NO,LTB4,IL-1β in the inflammatory reaction.
