小青龙汤合香砂六君子汤对过敏性哮喘小鼠气道反应性与细胞因子的影响

Effects of Xiao-Qing-Long Decoction Combined with Xiang-Sha-Liu-Jun-Zi Decoction on Airway Hyperresponsiveness and Cytokine Levels in Murine Model of Allergic Asthma

目的:探讨中药名方小青龙汤与香砂六君子汤的合方,对于小鼠过敏性哮喘模型气道反应性与细胞因子的影响。方法:将20只雌性BALB/c小鼠,随机平均分为正常对照组(control组)、哮喘模型组(OVA组)、中药合方低剂量组(LD组)、中药合方高剂量组(HD组)。OVA、LD、HD组使用卵清蛋白(OVA)腹腔注射致敏与雾化吸入激发以制作哮喘模型。在OVA激发结束后24 h以整体体积描记法检测小鼠气道反应性(Penh)。将小鼠犠牲后,收集支气管肺泡灌洗液(BALF),计算嗜酸性颗粒细胞(EOS)所占比例,并将BALF经离心后取上清液,以酶联免疫吸附法(ELISA)测定其中白细胞介素-4,5,13(IL-4,IL-5,IL-13)与Ig E之含量变化。接着取小鼠肺叶支气管组织切片经HE染色后,以光学显微镜观察组织型态与炎性细胞浸润情况。结果:中药合方高、低剂量组之小鼠气道反应性(Penh)、BALF中EOS所占比例、BALF上清液中细胞因子(IL-4,IL-5,IL-13,Ig E)含量,与OVA组相比,均有显著下降(P<0.05)。结论:小青龙汤合香砂六君子汤能有效降低哮喘小鼠的气道反应性,调控细胞因子含量水平,从而改善哮喘之气道炎性反应。

Objective： To evaluate the effects of Xiao-qing-long decoction combined with Xiang-sha-liu-jun-zi decoction on airway hyper-responsiveness and cytokine levels in asthmatic murine model. Methods： Twenty female BALB / c mice were divided equally and rando m Ly into four groups,including normal control group（ control group）,asthmatic model group（ OVA group）,low dose Chinese medicine group（ LD group）,and high dose Chinese medicine group（ HD group）. Mice in OVA, LD, and HD groups were injected with ovalbumin（ OVA） intra-peritoneally（ i. p） and inhaled aerosol to finish sensitization and challenge. After sacrifice,the lungs and tracheas were removed from mice,and each tissue specimen was sliced into 6-μm sections,and was stained with HE method for light microscopic inspection. Results： Compared with OVA group,airway hyper-responsiveness（ Penh）,percentage of EOS and cytokine（ IL-4,IL-5,IL-13,Ig E） levels in BALF in both Chinese medicine groups were significantly reduced（ P 0. 05）. Conclusion： Oral administration of Xiao-qing-long decoction combined with Xiang-sha-liu-jun-zi decoction may remarkably reduce airway hyper-responsiveness and Th2 cytokine levels in asthmatic murine model. Although detailed mechanism is to be investigated,we assume that this Chinese medicine decoction may exert anti-asthmatic effects by immune-modulatory effects of inhibiting Th2 responses on the basis of our limited results.
