摘要
目的:研究比较不同试剂处理对菲牛蛭冻干粉抗凝血活性的影响。方法:采用凝血酶直接滴定法对不同试剂处理后的菲牛蛭冻干粉的抗凝血活性进行测定。结果:与对照组相比较,乙酸乙酯、正己烷、环己烷、丙酮、石油醚、蒸馏水6种试剂对菲牛蛭冻干粉抗凝血活性基本无影响;甲醇和乙醇对其活性损失最大,超过33%;氯仿、二氯甲烷损失较小,接近17%。结论:菲牛蛭冻干粉抗凝血活性强弱与其处理的试剂有较大关系。在菲牛蛭深加工时,建议选择石油醚、乙酸乙酯、蒸馏水3种试剂,既可保证其安全性,也能有效保留菲牛蛭冻干粉的抗凝血活性,最大程度利用资源。
Objective: To study on the anti-thrombin activity of Poecilobdella Manillensis powder dried with vacuum freeze drying technology with different reagents treatment. Methods: The anti-thrombin activities of these powders with different reagents treatment were measured by the direct thrombin titration. Results: Compared with the blank group,these powders respectively treated by ethyl acetate,acetone,cyclohexane,n-hexane,petroleum ether and distilled water almost have the same anti-thrombin activity. On the contrary,those powders with methanol and ethanol have the largest losses; the data is more than 33%. In addition,the group of chloroform or dichloromethane losses was close to 17%. Conclusion: The anti-thrombin activity of Poecilobdella manillensis freezing dried powder is strongly affected by reagents. The choice can guarantee the anti-thrombin activity and more security is using reagents like petroleum ether,ethyl acetate and distilled water. Furthermore,this can be the greatest degree of resource utilization.
