摘要
目的探讨急性心肌梗死大鼠心脏结区组织中连接蛋白(Cx30.2、Cx45)的表达。方法结扎大鼠冠状动脉左前降支建立急性心肌梗死模型,随机分为模型组(MI组,n=10)和假手术组(只穿线不结扎,SH组,n=10)。术后4周取心脏标本,采用免疫组化和逆转录聚合酶链式反应(RT-PCR)法检测心脏结区组织中Cx30.2和Cx45蛋白及mRNA的表达。结果术后4周MI组Cx30.2和Cx45蛋白表达增加,分布不均,无规则,与SH组相比,蛋白表达差异有高度统计学意义(P=0.00);Cx30.2和Cx45 mRNA在MI组的表达量亦增加,与SH组相比,差异有高度统计学意义(P=0.00);Cx30.2和Cx45 mRNA的表达具有明显正相关(r=0.938,P<0.05)。结论急性心肌梗死后心脏结区组织中Cx30.2和Cx45表达增加,二者mRNA表达呈正相关。
Objective To explore the expression of connexin30.2(Cx30.2) and connexin45(Cx45) in heart node area tissues of rats with acute myocardial infarction. Methods Via the ligation of the left coronary artery on rats, acute myocardial infarction models were built and randomly divided into two groups: the acute myocardial infarction model group(group MI, n = 10) and the sham-operation group(group SH, n = 10). Heart samples were extracted from rats 4 weeks after operation. The expression of Cx30.2 and Cx45 protein and m RNA in heart node area tissues was detected by immunohistochemistry and RT-PCR respectively. Results 4 weeks after operation, the expression of Cx30.2 and Cx45 protein increased in group MI, but scattered in disorder, compared with group SH, the differences of protein expression were statistically significant(P = 0.00). The expression of Cx30.2 and Cx45 mRNA increased in group MI,compared with group SH, the differences were statistically significant(P = 0.00). The expression of Cx30.2 mRNA was positively correlated with Cx45 m RNA(r = 0.938, P 〈0.05). Conclusion The expression of Cx30.2 and Cx45 protein and mRNA in heart node area tissues increase after acute myocardial infarction, and they are positively correlated.
