1个水稻不闭颖突变体(open hull 1)的鉴定与基因定位

摘要 水稻颖花的开放与闭合是个复杂的过程,其分子调控路径及作用机制尚不清楚。在水稻品种9311的辐射突变体库中发现了一份花器官变异突变体,主要表现为突变体小花开放后不闭合,暂命名为不闭颖突变体(open hull 1,oph1(t))。通过对突变体的性状调查、小花解剖观察、遗传分析,发现:突变体开花后,颖壳不闭合,结实率明显降低,其他农艺性状则无显著差异;突变体在开花时浆片与野生型无差异,但开花后一段时间内仍能保持膨胀状态。以oph1(t)/9516的F_2、F_3群体内突变体单株作为定位群体,最终将OPH1基因定位于第6染色体短臂,位于标记e55和e8之间,遗传距离为1.29 cM,是一个尚未报道的新基因。 The genetic mechanisms of rice floret opening and closing are still unclear.A mutant with open-hull after flowering was obtained from the mutant library of indica rice variety 93-11.The mutant was temporarily named as openhull 1(ophl(t)).The morphological investigation showed that ophl mutant exhibited unclosed glumes after flowering and the low seed setting.Moreover,the other agronomic traits of ophl were not changed when compared to its wild type.Anatomical analysis indicated that,the morphology of lodicules in ophl mutant was same as that of wild type 93-11 when flowering,but they can maintain plump for longer time after flowering,however,the lodicules in wild type only can maintain plump for shorter time and then disintegrate.The genetic analysis indicated that the mutant was controlled by a single recessive,and consequently OPH1(t) was mapped to the short arm of chromosome 6,between the STS marker e55 and e8,the genetic distances were 1.29 cM,respectively.Our results laid a solid foundation for further functional analysis of OPH1(t) in rice flower organ morphogenesis.
