摘要
【目的】研究不同灭菌条件对蓝果忍冬组培苗成活率的影响,筛选适宜蓝果忍冬组培苗生长的灭菌流程和方法,为批量生产蓝果忍冬组培苗提供参考。【方法】以蓝果忍冬蓓蕾、长白山1号、阿尔泰1号、蓝鸟4个不同品系冬眠枝条萌发芽(芽尖)为外植体,用75%酒精、0.1%HgCl2和2%NaClO进行不同消毒剂组合及不同消毒时间灭菌处理。【结果】单独使用75%酒精消毒30 s、0.1%HgCl2消毒时间≤6 min消毒效果较好,2%NaClO灭菌时间不宜超过6 min。75%酒精(30 s)+0.1%HgCl2(6 min)处理蓓蕾蓝果忍冬成活率最高,长白山1号蓝果忍冬对75%酒精反应敏感,75%酒精消毒可提高其外植体成活率,2%NaClO对阿尔泰1号蓝果忍冬有明显的消毒效果,而蓝鸟蓝果忍冬对2%NaClO反应不敏感,2%NaClO消毒对其外植体成活率没有明显影响。综合污染率、成活率等因素,不同品系的蓝果忍冬采用75%酒精(30 s)+0.1%HgCl2(6 min)灭菌效果较好。【结论】蓝果忍冬芽尖外植体最佳灭菌流程为:75%酒精(30 s)→3次无菌水冲洗→0.1%HgCl2(6 min)→6次无菌水冲洗。
【Objective】The effects of different sterilization conditions on the survival rate of blue honeysuckle were studied to select appropriate sterilization method and provide references for mass production of blue honeysuckle plantlets. 【Method】 The explants were sampled from four hibernation adorable sprouting branches of "Berel", "Changbaishan 1", "Aertai 1", and "Bluebird", respectively. They were sterilized by 75% alcohol, 2% NaClO and 0.1% HgCl2in different combinations and sterilization time. 【Result】The results showed that the sterilization time of 75% alcohol should be under 30 s. The sterilization with 0.1% HgCl2and 2% NaClO should be under 6 min. The sterilization with 75%(30 s) + 0.1% HgCl2(6 min) was suitable for "Bluebird", while 75% alcohol was suitable for "Changbaishan 1", because it’s explants survival rate was improved. The sterilization with 2% NaClO was suitable for "Aertai 1" but not "Bluebird". Comprehensively considering different sterilization conditions, 75%(30 s)+ 0.1% HgCl2(6 min) was most proper for different strains of blue honeysuckle and the sterilization effect was the best. 【Conclusion】The best sterilization method were 75% alcohol(30 s) → three times of sterile water flushing → 0.1% mercuric chloride(6 min) → six times of sterile water flush.
