摘要
目的:探讨姜黄素对缺氧复氧的乳鼠心肌细胞是否具有保护作用,这种保护作用是否与热休克蛋白70(HSP70)表达有关。方法:原代培养乳鼠心肌细胞,MTT法测IC50确定姜黄素浓度,建立缺氧复氧模型,分别测定各组细胞乳酸脱氢酶(LDH)活性、丙二醛(MDA)含量,缺氧/复氧细胞凋亡与坏死,并用免疫组织化学检测HSP70表达。结果:缺氧/复氧模型预先加姜黄素组较I/R组LDH活性与MDA含量下降,HSP70表达增加,细胞凋亡与坏死少。结论:姜黄素对体外培养的缺氧复氧心肌细胞具有保护和抗凋亡作用,姜黄素对缺氧复氧心肌细胞保护作用可能与HSP70表达增加有关。
Objective: to explore the protection of curcumin on rat cardiac myocytes processed by hypoxia-reoxygenation and whether the protective effect is related to the expression of heat shock protein 70( HSP70). Methods: Rat cardiac myocytes were cultured primarily. MTT assay was applied to measure the IC50 of curcumin. Hypoxia-reoxygenation model was set up. The activity of lactate dehydrogenase( LDH) and the content of malondialdehyde( MDA) from each group were measured respectively. The apoptosis and necrosis of cells processed by hypoxia-reoxygenation were detected and the expression of HSP70 with immune histochemical method. Results: Compared with I / R group,the activity of LDH and the content of MDA of the concentrations of curcumin group decreased,but the expression of HSP70 increased. At the same time the rate of apoptosis and necrosis was reduced. Conclusions: Curcumin has protection and antiapoptotic effect on rat cardiac myocytes cultured in vitro and processed by hypoxia-reoxygenation. The protective effect is related to the increased expression of HSP70.
