反相高效液相色谱法同时测定栀子金花丸中栀子苷、小檗碱、黄芩苷、大黄酸、大黄素、大黄酚的含量 被引量:20

摘要 目的建立反相高效液相色谱法同时测定栀子金花丸中栀子苷、小檗碱、黄芩苷、大黄酸、大黄素、大黄酚6种主要成分的含量。方法色谱条件为Gemini NX C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱,洗脱程序如下:0-10 min(7%A),10-45 min(7%-25.5%A),45-75 min(25.5%-64%A),75-90 min(7%A);检测波长:254 nm;流速:1.0m L·min^-1。结果栀子苷、小檗碱、黄芩苷、大黄酸、大黄素、大黄酚浓度分别在3.440-20.64、4.020-24.12、19.45-116.70、1.074-6.444、1.146-6.876、1.366-8.196μg·m L^-1与峰面积呈良好线性关系,平均回收率(n=6)分别为99.9%、98.8%、101.0%、99.6%、101.3%、98.1%,RSD分别为1.7%、1.8%、1.4%、1.7%、1.4%、0.9%。结论该方法简便、准确、重复性好,可用于栀子金花丸的质量控制。 Objective To establish a RP-HPLC method to determine geniposide, berberine, baicalin, rhein, emodin, chrysophanol in Zhizi Jinhua pills at the same time. Methods The method was performed on a C18 chromatographic column(Gemini NX, 250 mm × 4.6 mm, 5 μm), with acetonitrile(A)- 0.1%phosphoric acid(B) as the mobile phase in gradient elution. The elution profile was: 0- 10 min(7%A), 10- 45 min(7%- 25.5%A), 45- 75 min(25.5%- 64%A), 75- 90 min(7%A). The detective wavelength was 254 nm and the flow rate was 1.0 m L·min^-1. Results There were a good linearity between the concentration and peak area value when the concentrations of geniposide, berberine, baicalin, rhein, emodin, and chrysophanol were at 3.440- 20.64, 4.020- 24.12, 19.45-116.70, 1.074- 6.444, 1.146- 6.876, 1.366- 8.196 μg·m L^-1. The average recoveries(n = 6) of 6 components were 99.9%, 98.8%, 101.0%, 99.6%, 101.3% and 98.1%, with RSDs of 1.7%, 1.8%, 1.4%, 1.7%, 1.4% and 0.9%, respectivly. Conclusion The method is simple, accurtate and reproducible. It can be used for the quality control of Zhizi Jinhua pills.
