双酶分步水解制备鱿鱼皮胶原蛋白肽的工艺条件优化 被引量:8

摘要 通过单因素试验和正交试验对鱿鱼皮胶原蛋白肽的双酶分步提取工艺进行优化。碱性蛋白酶与木瓜蛋白酶分步提取的最佳工艺条件:先用0.3%碱性蛋白酶于p H10、50℃条件下水解4 h,再用0.2%木瓜蛋白酶于p H8、55℃条件下水解2 h。在此最佳水解条件下得到的鱿鱼皮胶原蛋白肽水解度为24.32%,DPPH清除率为90.34%,多肽转化率为58.68%。采用SDS-PAGE电泳初步分析所得胶原蛋白肽分子量范围,结果表明,其分子量基本小于3000 u。 One-factor-at-a-time and orthogonal array methods were used to investigate the optimization of stepwise dual-enzymatic preparation of collagen peptides from squid skin in this work. The optimum extraction conditions of alcalase and papain: firstly, Alacalase hydrolyzed for 3 h under the conditions of 50 ℃, pH 10.0 and enzyme concentration 0.3%, then papain hydrolyzed for 2 h under the conditions of 55 ℃, pH 8.0 and enzyme concentration 0.2%. Under these condition, the maximum DH was 24.32%, the DPPH scavenging rate was 90.34% and recovery ratio of peptides was 58.68%. SDS-PAGE was used to analysis the molecular weight range of collagen peptide, result showed the molecular weight mostly less than 3000 u.
