摘要
目的探讨枸杞多糖(LBP)对糖尿病肾病兔肾脏的保护作用及单核细胞趋化蛋白-1(MCP-1)、细胞间粘附分子-1(ICAM-1)m RNA表达的影响。方法采用四氧嘧啶(ALX)诱导糖尿病肾病兔动物模型。将20只雄性日本大耳白兔随机分为5组,每组4只,分别为正常对照组、DN模型组、LBP预防组、LBP治疗组和阳性对照组。LBP预防组于糖尿病(DM)成模后即灌胃LBP(10mg/kg)持续12w,同时正常组与DN模型组分别灌胃等体积的生理盐水。LBP治疗组和阳性组于糖尿病肾病(DN)成模(第8w)后分别灌胃LBP(10mg/kg)和美卡素(3.7mg/kg)持续4w。实验第4、6、8和12w测空腹血糖及24h尿蛋白定量;实验第12w,采集血液标本,测定C反应蛋白(CRP)、尿素氮(BUN)和血肌酐(SCr),RT-PCR方法测定肾皮质MCP-1、ICAM-1m RNA表达。结果与正常组相比,DN模型组大耳白兔24h尿蛋白明显升高(P<0.01),血清中BUN、SCr和CRP含量明显增加(P<0.01),肾皮质MCP-1、ICAM-1m RNA表达明显上升(P<0.01)。LBP预防组大耳白兔24h尿蛋白明显降低,血清中BUN、SCr和CRP明显降低,肾皮质MCP-1、ICAM-1m RNA表达明显下调,与DN模型组比较均有显著性差异(P<0.01)。LBP治疗组与DN模型组相比,24h尿蛋白明显下降(P<0.05),血清中BUN、CRP明显降低(P<0.01),肾皮质MCP-1、ICAM-1m RNA表达明显降低(P<0.01)。结论 LBP能改善DN兔肾功能,减少MCP-1、ICAM-1m RNA表达,减轻炎症反应,延缓DN的发生发展。
Objective To investigate the effect of Lycium barbarum polysaccharides(LBP) on renal function and expression of MCP-1 mRNA and ICAM-1 mRNA in diabetic nephropathy rabbits. Methods Diabetes was induced by injecting alloxan(ALX). Japanese white rabbits were randomly divided into 5 groups: normal control group, diabetic nephropathy(DN)model group, LBP prevention group, LBP treatment group and positive control group(n=4 for each group). LBP(10 mg/kg) was given to LBP prevention group after diabetes mellitus(DM) model was successfully established for 12 weeks and to LBP treatment group after DN model was successfully established for 4 w. Telmisartan(3.7 mg/kg) was given to positive group after DN model was induced successfulty for 4 w. Normal group and DN group were given balanced saline. Fasting blood glucose and 24 h urine protein were detected at 4w, 6w, 8w and 12 w, and serum C-reactive protein(CRP) blood, urea nitrogen(BUN) and serum creatinine(SCr) were detected at 12 w. The expression of MCP-1 mRNA and ICAM-1 mRNA extracted from cortex were detected by RT-PCR. Result Twenty four hours urine protein was significantly increased in DN group compared with normal group(P〈0.01), and serum BUN, SCr and CRP were also increased(P〈0.01). The expression of MCP-1 mRNA and ICAM-1 mRNA from cortex were increased(P〈0.01). Compared with DN group, 24 h urine protein, BUN, SCr and CRP, the expression of MCP-1mRNA and ICAM-1mRNA were significantly decreased in LBP prevention group and LBP treatment group(P〈0.05, P〈0.01). Conclusion LBP can improve renal function of DN rabbits and reduce the expression of MCP-1 mRNA and ICAM-1 mRNA and delay the development of DN.
