人参、黄芩仿生化提取物对UVB照射HaCaT细胞HIF-1α表达的影响 被引量:4

摘要 目的探讨人参、黄芩仿生化提取物(SQBE)对中波紫外线(UVB)照射人永生化角质形成细胞(Ha Ca T)缺氧诱导因子1α(HIF-1α)表达的影响,阐明SQBE预防UVB辐射所致皮肤癌发生的可能作用机制。方法将对数生长期的Ha Ca T细胞,接种于含有10%胎牛血清DMEM培养基的培养皿中,于37℃、5%CO2的细胞培养箱内常规培养;分为对照组和SQBE 1组(终浓度为12.5μg·ml-1的SQBE)、SQBE 2组(终浓度为25.0μg·ml-1的SQBE)、SQBE 3组(终浓度为50.0μg·ml-1的SQBE),各组在给药4 h后与对照组同时进行UVB(0、20、40、60 m J·cm-2)照射;继续培养24 h后,采用MTT法检测细胞生存率,荧光定量PCR法及Western blotting法检测HIF-1αmRNA及HIF-1α蛋白的表达。结果与对照组相同照射剂量比较,SQBE 3组的细胞生存率明显提高(P<0.05),SQBE1组和SQBE 2组差异无统计学意义(P>0.05);SQBE 3组能够有效降低细胞内HIF-1αmRNA及HIF-1α蛋白的表达。结论大剂量人参、黄芩仿生化提取物(SQBE)可通过对细胞内HIF-1α表达的影响,预防UVB辐射导致的皮肤癌发生。 Objective To discuss the effect of ginseng and scutellaria bionic extract( SQBE) on HIF-1α expression in Ha Ca T cells irradiated by UVB and to expound the possible mechanism of anti-UVB radiation on skin cancer. Methods Logarithmic growth phase Ha Ca T cells were seeded in petri dishes containing10% fetal bovine serum in DMEM medium,and cultured in 37 ℃,5% CO2incubator; We divided into a control group,SQBE 1 group( 12. 5 μg · ml- 1SQBE),SQBE 2 group( 25. 0 μg·ml- 1SQBE),and SQBE 3 group( 50. 0 μg·ml- 1SQBE),each group was irradiated by UVB( 0,20,40,60 m J·cm- 2)4 hours after administration. After 24 hours,the cell survival rate was detected by MTT method; expression of HIF-1αmRNA and HIF-1α protein were detected by real-time PCR and Western blotting. Results Compared with the control group at the same dose,the cell survival rate in the SQBE 3 group was increased statistically( P〈0. 05),and no significant difference was found between SQBE 1 group and SQBE 2 group( P〈0. 05); the expressions of HIF-1αmRNA and HIF-1α protein were decreased markedly in the SQBE3 group. Conclusion SQBE can prevent the UVB-induced skin cancer by influcing the expression of HIF-1α in cells possibly.
