摘要
为了研究白腐真菌对黄芩苷生物的转化方法和模型,采用生长细胞转化法和静态细胞转化法对黄芩苷进行生物转化,使用超声波处理培养液,利用高效液相色谱检测转化产物.检测结果显示,在黄芩素标准品相应保留时间内有明显出峰,表明白腐真菌内的诱导酶、胞内酶能将黄芩苷转化为黄芩素.经实验确定最佳转化时间为96h,最佳pH为5.5,当料液比为1∶100时,最佳接种量是4片直径为10mm的菌饼.
The method and the model of biotransformation process of baicalin by white-rot fungi were studied.The dynamic cells and static cells were used to transform baicalin.After ultrasonic treatment on cultivation medium,the product was identified as the baicalein by HPLC.As a result,the transformation enzyme is supposed to be induced enzyme and endoenzyme at the same time.The optimal authire conditions are as follows:the reaction time is 96 h,pH is 5.5,inoculation amount is 4slices of white-rot fungi whose diameter is 10mm。
