摘要 优化燕麦成熟胚再生体系,缩短再生培养时间,为燕麦成熟胚转化体系的建立奠定基础。以燕麦品燕1号成熟胚为试验材料,采用全胚乳和无胚乳2种方式进行外植体脱分化培养。结果表明,无胚乳处理形成的愈伤组织质地最佳,胚性强;IM2(MS+3 mg/L 2,4-D+1 mg/L 6-BA+1 mg/L NAA+500 mg/L CH+500 mg/L Pro)芽分化诱导培养基对愈伤组织的再分化效果最佳;不同基因型的愈伤组织在IM2芽分化诱导培养基上诱导能力差异较大。 Optimization of regeneration system of mature embryo of oat, shorten the regeneration culture time, laid the foundation for the establishment of oats mature embryo transformation system. The experiment material was Pinyan 1 mature embryo. The experiment used the entire endosperm and without endosperm treatment conduct two kinds of explants dedifferentiation culture, in which without endosperm treatment formed callus texture best, embrygenic strong. The formation of callus in IM1, IM2, IM3 and IMd medium were cultured, the IM2(MS + 3 mg/L 2,4-D + 1 mg/L 6-BA + 1 mg/L NAA + 500 mg/L CH + 500 mg/L Pro) medium on callus diffe^ntiation was the best. To investigate the effect of different genotypes on the in vitro culture of mature embryo of oat from the use of 10 oat varieties, the results showed that different genotypes in IM2 differentiation medium effect on the differentiation of large differences.
