摘要
目的探讨周期素依赖性蛋白激酶5(Cdk5)和血管内皮生长因子(VEGF)在糖尿病肾病肾小球足细胞中的表达及相关性。方法选用db/db小鼠作为实验组(n=5),C57BL/6J小鼠作为对照组(n=5),2组小鼠在相同的饲养条件下,饲养至第14周并将其处死,取其肾脏,组织切片,采用免疫组化染色法测定Cdk5和VEGF表达并半定量分析。结果实验组、对照组小鼠Cdk5的表达均主要沿肾小球基底膜及足细胞分布,但实验组的表达强度高于对照组(实验组MOD值0.510±0.024,对照组MOD值0.486±0.023,P<0.05);对照组小鼠VEGF仅在足细胞有较弱表达(MOD值0.249±0.015 3),而实验组VEGF除了在肾小球足细胞阳性表达外,还可见系膜区阳性表达且表达强度明显增加(MOD值0.276±0.020),差异有统计学意义(P<0.05);Cdk5与VEGF表达的MOD值呈正相关(r=0.99)。结论 Cdk5和VEGF在糖尿病肾病肾小球足细胞中表达均增加,两者呈正相关关系,提示Cdk5和VEGF可能参与DN的发生发展。
Objective To discuss the expression and connection of Cdk5 and VEGF in the glomeruler podocytes of diabetic nephropathy. Methods In this study,selected db / db mice as experimental groups( n = 5),and C57 BL /6J mice as controls( n = 5). Both of them were feed under the same conditions and enviroment. They were sacrificed at 14 th week. Then the kidneys were dehydrated and embedded in paraffin,the tissue were sliced. The expressions of Cdk5 and VEGF were detected by immunohistochemical staining,and then data was analyzed. Results The expression of Cdk5 mainly distributed along the glomerular basement membrane and podocytes,whereas the expression intensity of Cdk5 in experimental mice was higher than that in the controls( the MOD value 0. 510 ± 0. 024 vs.0. 486 ± 0. 023,P 〈 0. 05); in the control group,VEGF mainly distributed in the glomerulus podocytes,however,in the DN group,VEGF distributed not only in the glomerulus podocytes,but also in mesangial area and its expression intensity is significantly increased comparing to the control group( the MOD value 0. 276 ± 0. 020 vs. 0. 249 ± 0. 015,P 〈 0. 05); The correlation analysis of Cdk5 and VEGF was positively correlated. Conclusion The expressions of Cdk5 and VEGF are increased in the glomerular podocytes of diabetic nephropathy,and both are positively correlated. It is suggested that Cdk5 and VEGF may be involved in the occurrence and development of DN process.
