c-Kit和PDGFR在皮肤肿瘤中的表达及意义

摘要 目的探讨酪氨酸蛋白激酶家族成员c-Kit和血小板衍生生长因子受体(PDGFR)在皮肤肿瘤患者中的表达,并分析其临床意义。方法选取本院2006年5月至2012年5月收治的152例皮肤肿瘤标本,良性肿瘤10例(良性组);30例为恶性肿瘤(恶性组)。并选取同期10例皮肤正常组织(正常组)。结果恶性组c-Kit突变阴性2例,阳性28例,阳性率93.3%;良性组阴性9例,阳性1例,阳性率10.0%。恶性组c-Kit阳性率显著高于良性组(2=6.515,P〈0.05)。恶性组PDGFR突变阴性3例,阳性27例,阳性率90.0%;良性组阴性9例,阳性1例,阳性率10.0%。恶性组PDGFR阳性率显著高于良性组(χ~2=7.609,P〈0.05)。恶性组DNA含量及PGDFR荧光强度DI为(1.248±0.198),FI为(1.509±0.377),显著高于良性组及正常组(P〈0.05)。结论 c-Kit和PDGFR基因突变在皮肤肿瘤,尤其是恶性肿瘤的发病中具有重要意义,其与皮肤肿瘤的发病具有较高的相关性,早期进行标本c-Kit和PDGFR基因检测,可帮助判断肿瘤分化、浸润程度。 Objective To explore the expression and significance of the tyrosine protein kinase family member C-Kit and PDGFR in dermatoma.Methods 40 patients with dermatoma in our hospital from May 2006 to May 2012 were selected,including 10 cases with benign tumor(benign group) and 30 cases with cancer(cancer group).And 10 normal skin specimens were selected as the control group.Results There were 2 cKit negative cases and 28 c-Kit positive cases in the cancer group,with a positive rate of 93.3%.There were 9 c-Kit negative cases and 1 positive cKit case in the benign group,with a positive rate of 10.0%.The c-Kit positive rate of the cancer group was significantly higher than that of the benign group(P〈0.05).There were 3 PGDFR negative cases and 27 PGDFR positive cases in the cancer group,with a positive rate of 90.0%.There were 9PGDFR negative cases and 1 PGDFR positive case in the cancer group,with a positive rate of 10.0%.The PGDFR positive rate of the cancer group was significantly higher than that of the benign group(P〈0.05).The DNA content and fluorescence intensity of PGDFR in the cancer group was(1.248+0.198) and(1.509±0.377),which were significantly higher than those in the benign group and control group(P〈0.05).Conlusion The c-Kit and PDGFR gene mutations played an important role in the pathogenesis of skin tumor,especially the malignant tumor,and had high correlation with the incidence of skin cance.The early testing can help to determine the degree of tumor differentiation and infiltration.
