摘要
目的探讨高血压(HTN)对糖尿病周围神经病(DPN)大鼠模型坐骨神经中髓鞘相关糖蛋白(MAG)和髓鞘碱性蛋白(MBP)表达的影响。方法大鼠随机分为正常组、DPN组和DPN+HTN组。造模后4周检测各组血压、血糖及坐骨神经传导速度,RT-PCR检测坐骨神经MAG和MBPmRNA表达。结果与正常组比较,DPN组和HTN+DPN组血糖显著增高(P<0.01),神经传导速度显著降低(P<0.01),坐骨神经MBP和MAG mRNA表达显著上调(P<0.01),其中HTN+DPN组与DPN组比较收缩压显著增高(P<0.01),神经传导速度显著降低(P<0.01),坐骨神经MAG mRNA表达显著升高(P<0.01),MBP mRNA表达显著下降(P<0.01)。结论高血压能够上调MAG和下调MBP表达,抑制髓鞘形成,加剧糖尿病周围神经病的神经损伤。
Objective To investigate the effects of hypertension(HTN)on the expression of myelin associated glycoprotein (MAG) and myelin basic protein(MBP)in the sciatic nerve in rats with diabetic peripheral neuropathy(DPN).Methods The rats were randomly di-vided into normal group,DPN group and HTN+DPN group.After 4 weeks of modeling,the blood sugar,blood pressure and conduc-tion velocity of sciatic nerve were measured,the mRNA expression of MBP and MAG in the sciatic nerve was detected by RT PCR. Results After 4 weeks of modeling,compared with normal group,the blood sugar were increased in DPN group and HTN+DPN group,while the conduction velocity of sciatic nerve was decreased,the mRNA expression of MAG and MBP in sciatic nerve were in-creased (P〈0.01).Compared with DPN group,the blood pressure were increased in HTN+DPN group,and the conduction velocity of sciatic nerve was decreased,the mRNA expression of MAG was increased and the expression of MBP was decreased in sciatic nerve (P〈0.01).Conclusion Hypertension can contribute to worsening of DPN by elevating MAG and decreasing MBP in sciatic nerve,and inhabiting myelination.
