白藜芦醇抗大鼠心肌缺血再灌注损伤时心律失常的作用 被引量:2

摘要 目的:探讨白藜芦醇对大鼠心肌缺血再灌注损伤时心律失常的作用。方法:健康雄性SD大鼠40只,随机数字量表法分为4组(假手术组、缺血再灌注组、缺血预适应组、白藜芦醇组),采用结扎冠状动脉前降支制备心肌缺血再灌注损伤模型,于结扎前15min及再灌注前1min经舌下静脉注射白藜芦醇10mg/kg。BL420S生物信号采集处理系统记录肢体Ⅱ导联心电图变化及心律失常评分,记录左心功能指标LVSP、LVEDP、±dp/dtmax。TTC染色计算心肌梗死面积。测定血浆中CK-MB、MDA含量及SOD、GSH-Px活性。结果:白藜芦醇与缺血预适应组或缺血再灌注组相比,心律失常评分下降(P<0.01),心肌梗死面积缩小(P<0.01或P<0.05),LVSP提高、LVEDP下降、±dp/dtmax提高(P<0.05或P<0.01),CK-MB、MDA含量减少(P<0.05),SOD、GSH-Px活性增强(P<0.05或P<0.01)。结论:白藜芦醇可通过抗氧化作用防止心肌缺血再灌注时心律失常的发生,缩小心肌梗死面积,改善心脏舒缩功能。 Objective:The purpose of this study was to investigate the antiarrhythmic effects of resveratrol on rat during myocardial ischemia-reperfusion injury.Methods:Forty healthy male Sprague-Dawley(SD)rats were randomly divided into 4groups(sham operation group,ischemia reperfusion group,ischemic preconditioning group,resveratrol group).The myocardial ischemia reperfusion injury model was induced by ligating anterior descending branch of coronary artery,which was intravenous injection of resveratrol 10mg/kg before ligation of15 min and reperfusion of 1min.It was recorded for II-lead electrocardiogram,arrhythmias score and the left heart function indexes of left ventricular systolic pressure(LVSP),left ventricular end diastolic pressure(LVEDP),maximum rate of change of left ventricular pressure(±dp/dtmax)by BL420 Sbiological signal collecting and processing system.Myocardial infarction size was determined by TTC stain.It was measured for the creatinine kinase-MB(CK-MB)or malondialdehyde(MDA)contents and superoxide dismutase(SOD)or glutathione peroxidase(GSH-Px)activities in plasma.Results:Resveratrol can decrease the score of arrhythmia,reduce the area of myocardial infarction.Second,Resveratrol can improve the LVSP and±dp/dtmax,decrease the LVEDP.The above data changes were accompanied with the increase of SOD or GSH-Px activities and the decrease of CK-MB or MDA contents in plasma.Conclusion:Our datas suggest that resveratrol may prevent the occurrence of arrhythmia,reduce the area of myocardial infarction and improve cardiac systolic and diastolic functions through its antioxidant effect during myocardial ischemia reperfusion.
