A simple method for obtaining single spore isolates of Podosphaera xanthii

为了建立一种新的甜瓜白粉病病原菌单孢子分离方法,采用眉毛挑针挑取法,即在实体解剖镜下,在水琼脂培养基上直接挑取单个孢子进行活体分离纯培养。结果表明,此方法简单快速、操作方便、易于掌握,非常适用于孢子较大且活体寄生的病原菌。首次建立了甜瓜白粉病单个孢子活体分离纯培养的方法,为甜瓜白粉病生理小种及相关育种研究奠定了基础。

This research established a new method of eyebrows pick method for single spore isolation. The method was simple,rapid,convenient,and easy to use. The method is very suitable for larger spores and living parasitic pathogens. This method is first used for making pure culture of Podosphaera xanthii. The method laid the foundation for melon breeding for resistance to powdery mildew and for the study of pathogen physiological races.
