摘要
目的观察顺铂植入剂植入对人胃癌裸鼠移植瘤模型中肿瘤细胞凋亡以及脾酪氨酸激酶(SYK)、人表皮生长因子受体2(HER2)蛋白表达的影响,探讨其作用的相关机制。方法采用人胃癌SGC7901细胞系建立BALB/c裸鼠皮下移植瘤模型,随机分为A、B、C、D 4组,每组5只,A组尾静脉注射PBS,B组尾静脉注射顺铂,C组瘤内注射顺铂,D组瘤内植入顺铂植入剂。干预后,记录瘤重,计算抑瘤率,并对瘤体进行HE染色,通过免疫组化法检测SYK及HER2的表达,并取每组部分瘤体制成细胞混悬液进行流式细胞凋亡检测。结果 D组在抑制移植瘤生长、促进肿瘤细胞凋亡方面较其余3组更具有明显优势,且瘤组织SYK的阳性细胞率明显高于其余3组,而HER2的阳性细胞率在4组中为最低。结论顺铂植入剂可能通过下调HER2或上调SYK的表达来促进癌细胞的凋亡以发挥抗肿瘤的作用,从而为临床胃癌的治疗提供新思路。
Objective To observe the effects of sustained -release cisplatin implantation on tumor cell apoptosis and the SYK and HER2 protein expression in nude mouse with subcutaneous gastric carcinoma transplantation . Methods The subcutaneous gastric carcinoma transplantation model with SGC 7901 cell was constructed using BALB/c nude mice, which were further randomly divided into 4 groups:control group (received tail intravenous injection of PBS , Group A), Group B (received tail intravenous injection of cisplatin ), Group C (received intratumor injection of cispla-tin), and Group D (received the implant of cisplatin -implants).After the drugs intervening, the tumors were harvested for assessment of the tumor weight and tumor inhibiting rate , HE staining observation , examination of SYK and HER 2 expression , and assessment of cell apoptosis .Results Sustained-release cisplatin significantly promoted the apoptosis as well as suppressed the tumor growth , with significantly up -regulated the expression of SYK and down -regulated the expression of HER2.Conclusion Cisplatin-implants promotes the tumor apoptosis by enhancing the expression of SYK and reducing the expression of HER 2.
