摘要
目的探讨蒽贝素对家兔心脏骤停模型中心肌缺血再灌注损伤的保护作用。方法 30只成年雄性新西兰大白兔随机分为3组:假手术(Sham)组,CPR组和蒽贝素组。检测促炎因子(TNF-α,IL-1β及IL-6),核因子-κB(NF-κB)p65的表达水平,心脏肌钙蛋白(c Tn I),监测血流动力学、检测心肌坏死率、细胞凋亡指数(AI)以及组织学损伤程度。结果较Sham组,CPR组与蒽贝素组TNF-α,IL-1β及IL-6的表达水平均提高(P均<0.05)。较CPR组,蒽贝素组有效地下调了TNF-α,IL-1β及IL-6的表达水平(P<0.05),并下调了血清c Tn I表达水平,降低坏死率与细胞凋亡指数,下调了NF-κB p65表达水平(P<0.05)。同时,蒽贝素改善了血流动力学,心肌功能以及心肌形态。结论蒽贝素通过其抗炎作用有效的改善心脏骤停后心肌缺血再灌注损伤,从而起到保护心脏的作用。
ObjectiveTo investigate the protective effects of embelin on myocardial ischemia-reperfusion(IRI) injury following cardiac arrest (CA) in a rabbit model.MethodsThirty rabbits were randomly divided into three groups:sham group, CPR group, and embelin group. Pro-inflammatory (TNF-α, IL-1β, and IL-6) cytokines, nuclear factor-kappa B (NF-κB) p65, cardiactroponin I (cTnI), hemodynamics, necrosis ratio, apoptotic index (AI), and histological damage had been measured or evaluated.ResultsThe expressions of TNF-α, IL-1β, and IL-6 were significantly lower in embelin groups than those in the CPR group, but higher than those in the sham group (P〈0.05). Compared with the CPR group, embelin groups reduced the serum level of cTnI, the necrosis ratio, the AI, and the expression of NF-κB p65 (P〈0.05). Meanwhile, it improved the hemodynamics and myocardial function. Moreover, embelin-treated groups also showed improved myocardial morphology.ConclusionEmbelin may protect the heart against myocardial IRI following CA via its anti-inflammatory abilities.
