骨骼肌与心肌化学去细胞生物支架对大鼠脊髓半损伤脊髓修复的实验研究

摘要 目的探究与比较采用带血管蒂肌瓣贴附以及填充肌基膜和心肌,从而使得脊髓损伤缺血区的血液供应能力获得增强,同时使脊髓继发性损伤得以减弱的三种手术方法。方法本研究通过手术的方法,在Wistar大鼠脊髓损伤处的上方覆盖带蒂肌瓣,以及分别在脊髓损伤处填充肌基膜和心肌。然后,在手术结束后4周,对实验对象进行灌注处理,同时观察其组织学染色情况,最后使用电镜观察其轴突的变化情况。结果染色结果显示,进行带蒂肌瓣覆盖的Wistar大鼠的脊髓损伤处有血管生长,并且该血管从肌肉延伸到脊髓实质,另外进行肌基膜和心肌填充的Wistar大鼠同样出现有血管生成的现象,与此同时,电镜结果显示其都有神经轴突的生成,显示出Wistar大鼠的继发性损伤在很大程度上有所降低。结论三种方法均能够有效地加强血液在缺血区的供应,使继发性损伤程度降低,具有很好的临床应用前景。 Objective To explore and compare with vascular pedicle muscle flap attached,filled with myocardial muscle basement mem-brane,thus making the get enhance its capability of blood supply of spinal cord injury of ischemia area,at the same time make the spinal cord sec-ondary injury subsides three surgical methods. Methods By using the method of operation,cover in the upper part of the spinal cord lesions pedi-cle muscle flap,and in the spinal cord lesions filled membrane and cardiac muscle,respectively. Then,at 4 weeks after surgery,perfusion treat-ment of the subjects was observed,including the histological staining. Finally electron microscope was used to observe the changes of the axon. Results Dyeing experiment object showed pedicle muscle flap to cover the subjects had blood vessel growth of spinal cord injury,and the blood vessels from muscle extension to the spinal cord parenchyma,in addition to muscle membrane. The subjects of myocardial fill also appeared the phenomenon of angiogenesis. At the same time,the electron microscope results showed that the god located the place where they generate. This method showed that the subjects of secondary damage reduced to a great extent. Conclusion This paper demonstrated that three methods can ef-fectively strengthen the blood supply in the ischemic area,reduce secondary damage degree. The three methods had good clinical application pros-pects.
