摘要
目的:观察通脉大生片对大鼠卵巢TGF-β1、Smad7在TGF-β/smads通路中的表达变化及意义。方法:建立大鼠肾虚排卵障碍不孕(ASR)模型,通过阴道涂片筛选造模成功大鼠后分组用药,采用酶联免疫法(Elisa)法检测TGF-β1的表达,蛋白免疫印迹法(Western Blot)检测大鼠卵巢中Smad7蛋白的表达量。结果:①模型组大鼠卵巢TGF-β1的表达量最低,与正常组、通高组、来曲唑组、右归丸组相比,有统计学差异(P<0.05,P<0.01);通脉大生片组中以通高组最优;②通脉大生片高、中组来曲唑组及右归丸组Smad7表达均低于模型组(P<0.05,P<0.01),以通高组效果最好。结论:通脉大生片可用于治疗肾虚排卵功能障碍型不孕,而其促排卵作用可能部分是通过调节TGF-β/smads信号通路中TGF-β1及Smad7的表达,进而促进排卵障碍型不孕大鼠卵巢颗粒细胞TGF-β/smads信号通路的激活来实现的。
Objective: To observe the effects of Tongmai Dasheng Tablet on expression of TGF-β 1 and Smad7 in rat ovary. Methods: After establishing rat ovulatory obstacle sterility induced by kidney deficiency model, vaginal smear was used to screen the rats. The level of TGF-β1 was detected with Elisa method. The protein expression of Smad7 in ovary was detected with Western Blot method. Results: (1)Compared with the model group, the level of TGF-β 1 in blank control group increased, with statistical differences, as well as in high-dose Tongmai Dasheng Tablet group, letrozole group and Yougui Pill group (P〈0.05, P〈0.01). The level of TGF- β1 in high-dose Tongmai Dasheng Tablet group was the highest among all Tongmai Dasheng Tablet groups. (2)Compared with model group, the protein expression of Smad7 in high, middle dose Tongmai Dasheng Tablet group and letrozole group were all decreased (P〈0.05, P〈0.01). The protein expression of Smad7 in high-dose Tongmai Dasheng Tablet group was the lowest among all groups. Conclusion: Tongmai Dasheng Tablet could be used to treat ovulatory obstacle sterility with kidney deficiency syndrome, the effect of which may be partly related to activate the expressions of TGF-β and Smad7 in TGF- β/smads signaling pathway in ovarian granular cells.
