摘要
目的:通过观察中药清肠愈疡汤对溃疡性结肠炎模型大鼠一般生理状态、疾病活动指数(DAI)评分、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)产生的影响,探讨其作用机制。方法:将大鼠按体质量随机分为5组,分别为正常组、模型组、清肠组、香连组、5-ASA组,分组饲养12d后,进行DAI评分,ELISA法检测大鼠血清中IL-1β、IL-6的含量。结果:清肠组治疗后DAI评分第4天、第8天、第12天分别为1.75±0.59,0.83±0.31,0.59±0.30,为逐渐降低趋势;大鼠血清中IL-1β与IL-6水平显著下降,分别达到(12.51±0.63)ng/L,(53.72±4.46)ng/L,清肠组与5-ASA组、香连组、模型组比较,均有显著性差异(P<0.05)。结论:清肠愈疡汤可以显著改善清肠组大鼠症状,能够下调大鼠血清中IL-1β、IL-6的水平,减轻大鼠结肠组织的炎症反应,而起到治疗溃疡性结肠炎的作用。
Objective: Observation of the effects of QingchangYuyang Decoction on the general physiological state, disease activity index (DAI) score, IL-1β and IL-6 in serum in ulcerative colitis rat model to explore its mechanism. Methods: According to the bodyweight, the rats were randomly divided into 5 groups, including normal group, model group, Qingchang group, Xianglian group and 5-ASA group. DAI score and the concentrations of IL-I [3 and IL-6 in serum were determined after the rats were fed for 12 days. Results: After the treatment, DAI score was gradually decreased in Qingchang group, with the DAI score as 1.75±0.59, 0.83±0.31 and 0.59±0.30 at the 4th, 8th, 12th day respectively. The concentrations of IL-1β and IL-6 in rats serum were obviously reduced, with the concentrations of IL-1β and IL-6 up to (12.51±0.63)ng/L and (53.72±4.46)ng/L respectively. Compared with Qingchang group, the concentrations of IL-1β and IL-6 in 5-ASA group, Xianglian group and model group, were significant differences (P〈0.05). Conclusion: Qinchang Yuyang Decoction could significantly improve the symptoms of ulcerative colitis in rats model, that may be related to down-regulation of the level of IL- 1β and IL-6 in serum and relieving the inflammation response in intestinal tissue, playing an important role of treatment for ulcerative colitis.
