miR-21对1型糖尿病患者外周血单个核细胞分泌IFN-γ与IL-17的影响 被引量:3

摘要 目的:探讨mi R-21在1型糖尿病患者外周血单个核细胞(PBMC)分泌干扰素-γ(IFN-γ)与白介素-17(IL-17)的影响。方法:采集60例1型糖尿病患者及60例正常健康人外周血,分离外周血单个核细胞(PBMC),用q RT-PCR检测两组PBMC中mi R-21的表达;向PBMC中转染mi R-21,ELISA检测PBMC分泌IFN-γ与IL-17情况。结果:1型糖尿病患者PBMC中mi R-21表达低于对照组,差异有统计学意义(P<0.01);增加1型糖尿病患者PBMC中mi R-21的表达,IFN-γ与IL-17的分泌均减少,差异有统计学意义(P值分别<0.05或<0.01)。结论:mi R-21在1型糖尿病患者PBMC中的表达下降,并可抑制PBMC中IFN-γ与IL-17的分泌。 Objective To investigate the effect of mi R-21 on the secretion of IFN-γ and IL-17 from peripheral blood mononuclear cells(PBMC) in type 1 diabetic patients(T1D). Methods The expression levels of mi R-21 in PBMC from 60 T1 D patients and controls by quantitative real-time quantitative polymerase chain reaction(q RT-PCR) were detected and the secretions of IFN-γ and IL-17 from T1 D PBMC after transfection with mi R-21 were detected by enzyme linked immunosorbent assay. Results The expression of mi R-21 in PBMC from T1 D patients was significantly lower than the control(P〈0.01). The levels of IFN-γ and IL-17 from T1 D PBMC after transfection with mi R-21 were remarkably downregulated compared to the control(P〈0.05 or P〈0.01).Conclusion Compared with that at the healthy control group, the expression of mi R-21 in PBMC from T1 D patients is decreased. Mi R-21 could suppress the secretions of IFN-γ and IL-17 from PBMC in T1 D patients.
