摘要 [目的]研究黄金艾蒿、香蒿、青蒿和艾蒿4种蒿属植物离体再生技术。[方法]以黄金艾蒿、香蒿的茎段和青蒿、艾蒿的花蕾为外植体,研究了不同植物生长调节剂和添加物对4种蒿属植物愈伤组织诱导、不定芽再生、增殖培养和生根培养4个阶段的影响。[结果]愈伤组织诱导阶段各蒿属植物的最佳培养基为:黄金艾蒿茎段MS+0.1~0.5mg·L^-1NAA+1.0mg·L^-16-BA,香蒿茎段MS+0.5mg·L^-1NAA+2.0mg·L^-16-BA,青蒿花蕾MS+0.5mg·L^-1NAA+1.0mg·L^-16-BA,艾蒿花蕾MS+0.5mg·L^-1NAA+1.0mg·L^-16-BA。不定芽再生阶段各蒿属植物的最佳培养基为:黄金艾蒿和香蒿MS+0.1mg·L^-1NAA+3.0mg·L^-16-BA,青蒿和艾蒿MS+0.5mg·L^-1NAA+1.0mg·L^-16-BA。增殖培养阶段各蒿属植物的最佳培养基为:黄金艾蒿和香蒿MS+0.3mg·L^-1IBA+0.5mg·L^-16-BA,青蒿MS+0.3mg·L^-1IBA+2.0mg·L^-16-BA,艾蒿MS+0.3mg·L^-1IBA+1.0mg·L^-16-BA。生根培养阶段,以1/2MS培养基为基本培养基,黄金艾蒿在IBA质量浓度为0mg·L^-1和活性炭质量浓度为1.0g·L^-1时表现最佳,香蒿在IBA为0.4mg·L^-1和活性炭为0.5g·L^-1时表现最佳,青蒿在IBA为0mg·L^-1和活性炭为0g·L^-1时表现最佳,艾蒿在IBA为0.4mg·L^-1和活性炭为0.5g·L^-1时表现最佳。[结论]各蒿属植物最适离体再生条件和再生效果受基因型差异影响明显,而活性炭对生根培养的作用具有两重性。 [Objectives]The regeneration technology of Artemisia vulgaris‘Variegate',A.annua,A.annua‘Belgium'and A.argyi was studied.[Methods]The stem of A.vulgaris‘Variegate'and A.annua,the bud of A.annua‘Belgium'and A.argyi were used as explants.The effects of different plant growth regulators and additives on four Artemisia plants were studied,including callus induction,shoot regeneration,shoot proliferation and rooting four stages.[Results]The results showed as follows: the best medium for callus induction stage was A.vulgaris‘Variegate'stem MS + 0.1-0.5mg·L^-1NAA + 1.0mg·L^-16-BA,A.annua stem MS + 0.5mg·L^-1NAA + 2.0mg·L^-16-BA,A.annua‘Belgium'bud MS + 0.5mg·L^-1NAA + 1.0mg·L^-16-BA,A.argyi bud MS + 0.5mg·L^-1NAA+ 1.0mg·L^-16-BA; the best medium for shoot regeneration stage was A.vulgaris‘Variegate'and A.annua MS + 0.1mg·L^-1NAA +3.0mg·L^-16-BA,A.annua‘Belgium'and A.argyi MS + 0.5mg·L^-1NAA + 1.0mg·L^-16-BA; the best medium for shoot proliferation stage was A.vulgaris‘Variegate'and A.annua MS + 0.3mg·L^-1IBA + 0.5mg·L^-16-BA,A.annua‘Belgium'MS + 0.3mg·L^-1IBA+ 2.0mg·L^-16-BA,A.argyi MS + 0.3mg·L^-1IBA + 1.0mg·L^-16-BA; at the rooting stage,using 1 /2 MS medium as basic medium,A.vulgaris‘Variegate'performed better when the concentration of IBA was 0mg·L^-1and the concentration of activated carbon was1.0 g·L^-1,A.annua performed better when the concentration of IBA was 0.4mg·L^-1and the concentration of activated carbon was 0.5 g·L^-1,A.annua‘Belgium'performed better when the concentration of IBA was 0mg·L^-1and the concentration of activated carbon was 0 g·L^-1,A.argyi performed better when the concentration of IBA was 0.4mg·L^-1and the concentration of activated carbon was0.5 g·L^-1.[Conclusions]The optimum conditions and effects of vitro regeneration of four Artemisia plants are apparently influenced by genotype differences,and the role of active carbon on rooting culture are diversified.
