摘要
目的探讨运动与共轭亚油酸(CLA)对青春期肥胖SD大鼠PPARγ的影响。方法采用雄性SD大鼠建立肥胖大鼠模型,选取建模成功大鼠32只,分为对照组、静止+CLA组、运动组、运动+CLA组,8周后采集大鼠血液及脂肪组织,测血脂,用qRT-PCR测脂肪组织中PPARγmRNA的表达量、免疫组化测脂肪组织PPARγ的蛋白表达,ELISA测血浆PPARγ浓度。结果 1运动组、静止+CLA组TC低于对照组(P<0.05);运动组、运动+CLA组TG水平低于对照组、静止+CLA组(P<0.01);运动组、运动+CLA组LDL-c高于对照组、静止+CLA组(P<0.05);HDL-c组间无差异。2运动组、运动+CLA组血浆PPARγ水平高于对照组和静止+CLA组(P<0.01);运动组、运动+CLA组脂肪组织PPARγmRNA的表达高于对照组和静止+CLA组(P<0.01),静止+CLA组血浆PPARγ浓度和脂肪组织mRNA表达高于对照组但无统计学意义。3PPARγ蛋白表达与PPARγmRNA的表达情况一致。结论 8周不同干预方法使青春期肥胖大鼠的血脂指标中TG、TC浓度降低,运动和运动+CLA降低TG优于单纯补充CLA。单纯运动和运动+CLA能提高大鼠脂肪组织中PPARγmRNA的表达和蛋白表达及血浆PPARγ的含量。
Objective To explore effects of exercise and conjugated linoleic acid( CLA) on PPARγ in adolescent obese male SD rats. Methods obese rats were modeled with high fat feeding,32 obese rats were selected and randomly divided into control group,static + CLA group,exercise-treated group,exercise + CLA treated group. Blood and adipose tissue were collected after 8 weeks,and blood lipid was measured. PPARγmRNA gene expression in adipose tissue was tested using qRT-PCR,PPARγ protein expression in adipose tissue by immunohistochemistry,concentration of PPARγ in plasma by ELISA. Results 1 TC level of static + CLA group and exercise group were lower than the control group( P〈0. 05). TG level of the exercise group and exercise group +CLA were lower than the control group,static + CLA group( P〈 0. 01). LDL-c level of exercise group and exercise + CLA group was higher than the control group( P〈 0. 05),HDL-c level have no difference in groups. 2 PPARγ concentration in plasma in exercise group,exercise + CLA group was higher than the control group and static + CLA group( P〈 0. 01). Expression of PPARγ mRNA in adipose tissue in exercise group,exercise +CLA group was higher than the control group and static + CLA group( P〈 0. 01). PPARγconcentration in plasma and expression of PPARγ mRNA in adipose tissue in static +CLA group were higher than control group but with no statistical significance. 3 The situation of PPARγ protein expression was the same with the expression of PPARγ mRNA.Conclusion 8 weeks of different intervention methods can reduce the concentration of TG and TC in blood lipid index in adolescent obese rats,TG of exercise and exercise + CLA is better decreasing than simply add CLA. Exercise and exercise + CLA can improve the expression of PPARγ mRNA and protein in adipose tissue and plasma PPARγconcentration in rats.
