摘要
目的制备兔组织笼无菌感染模型,为药物药动学-药效学结合模型(pharmacokinetic-pharmacodynamic modeling,PK-PD)研究提供在体动物实验载体。方法选取健康家兔28只,耳缘静脉注射盐酸赛拉嗪注射液0.15ml/kg麻醉,每只分别背部备皮、切开皮肤,然后在皮下植入长约5cm的无菌中空胸腔引流管1个,继而在无菌操作下手术缝合。术后应用青霉素肌肉注射3d以预防感染。结果 5只家兔先后因不同原因死亡,23只家兔均获得清亮的无菌组织液,成功建立兔组织模型,成功率为82.14%。结论通过严格控制无菌操作,选择合适的麻醉剂,手术刀口选择在兔背部,术后及时预防感染,可以成功制备兔组织笼无菌感染模型,并获得清亮的无菌组织液。
Objective To prepare a rabbit tissue cage model so as to provide drug animal carrier in vivo for drug PK-PD modeling study.Methods 28 healthy rabbits were selected and injected with the anesthesia xylazine iv respectively.The back hair was cut in each rabbit,and then the skin was cut open.A5 cm of sterile hollow chest tube drainage was implanted subcutaneously.In the end the wound was sewed up with aseptic technique.After operation penicillin im was injected for 3 days to prevent infection.Results Among the 28 rabbits,5 rabbits were dead due to different causes.Clear and sterile tissue fluid were gotten from other 23 rabbits.A rabbit tissue model was established successfully,and the success rate was 82.14%.Conclusion By stricted controlling aseptic operation,selecting the appropriate anesthesia and surgical incision in the back of the rabbit,timely postoperative anti-infection,a rabbit tissue cage model can be successfully established,so does clear tissue fluid.
