摘要
为比较玉竹多糖、桑叶多糖、杜仲多糖、商陆多糖、淫羊藿多糖体外增强免疫活性,采用酶学检测法测定各中药多糖对鸡胚成纤维细胞的安全浓度,根据安全浓度测定结果,选择250.000μg/ml、125.000μg/ml、62.500μg/ml、31.250μg/ml、15.625μg/ml 5个浓度,用MTT法比较5种中药多糖单独和PHA协同刺激对外周血T淋巴细胞增殖指数的影响,以及比较5种中药多糖单独和LPS协同刺激对脾脏B淋巴细胞增殖率的影响。结果显示,玉竹多糖、桑叶多糖、杜仲多糖、商陆多糖、淫羊藿多糖对鸡胚成纤维细胞的安全浓度分别为1 250.0μg/ml、625.0μg/ml、625.0μg/ml、625.0μg/ml、312.5μg/ml。玉竹多糖、桑叶多糖、杜仲多糖和淫羊藿多糖无论是单独还是协同PHA刺激外周血T淋巴细胞,均不同程度地表现出增强T淋巴细胞的活性。桑叶多糖、杜仲多糖和淫羊藿多糖无论是单独还是协同LPS刺激脾脏B淋巴细胞,均不同程度地显示出良好的增强B淋巴细胞的活性。桑叶多糖和杜仲多糖体外增强免疫效果最佳,在62.500μg/ml和125.000μg/ml时,均显示出较强的增强外周血T淋巴细胞和脾脏B淋巴细胞增殖的能力。
Polygonatum odoratum polysaccharide( POPS),morus polysaccharide( MPS),eucommia polysaccharide( EPS),pokeweed polysaccharide( PPS),and epimedium polysaccharide( EPPS) to chicken embryonic fibroblast( CEF) in vitro were determined and showed as follows compared. 1 250. 0 μg / ml,625. 0 μg / ml,625. 0 μg / ml,625. 0 μg / ml,and312. 5 μg / ml,respectively. The effects of above five polysaccharides applied solely or co-applied with phytohaema glutinin( PHA) on the proliferation of peripheral blood T lymphocytes and the effects of five paly saccharides appiied solely or co-applied with lipopolysaccharide LPS on the proliferation of spleenic B lymphocytes were compared. POPS,MPS,EPSand EPPScould enhance peripheral blood Tlymphocytes activity at different levels either stimulated alone or co-stimulated with PHA. MPS,EPSand EPPScould improve spleenic B lymphocytes activity at different levels either stimulated alone or co-stimulated with LPS. The immune enhancements of MPSand EPSin vitro outperformed others at the concentration of 62. 5 μg / ml and 125 μg / ml.
