Bcl-xL、Bcl-2与维吾尔族妇女宫颈癌关系的研究 被引量:5

Study on the relationship between Bcl- x L,Bcl- 2 and cervical cancer among Uygur women

摘要 目的:检测凋亡相关基因在维吾尔族妇女宫颈癌中的表达,探讨维吾尔族妇女宫颈癌的发病机制及其与临床病理指标的关系。方法:收集2012年6月-2013年3月新疆维吾尔自治区人民医院妇科活检及手术切除的子宫肌瘤正常宫颈组织标本21例及宫颈癌组织标本45例,采用实时荧光定量PCR及免疫组化法对宫颈癌组织及正常宫颈组织中Bcl-x L、Bcl-2进行检测,并分析其表达水平与相应临床病理指标的关系。结果:Bcl-x L在宫颈癌组织中表达上调,与患者的肿瘤直径大小、病理分级、临床分期、肌层浸润深度及有无淋巴结转移有关。Bcl-x L mRNA在宫颈癌组织中的表达量是在正常宫颈组织中表达量的4.68倍,Bcl-2 mRNA在宫颈癌组织中的表达量低于在正常宫颈组织中的表达量。Bcl-x L、Bcl-2蛋白在宫颈癌组织和正常宫颈组织中的阳性表达率比较,差异均有统计学意义(χ2=12.357,P=0.000;χ2=5.820,P=0.015)。在宫颈癌组织中,Bcl-x L、Bcl-2蛋白在不同临床分期、肿瘤直径、临床分期、肌层浸润深度、病理分级及有无淋巴结转移中的表达水平比较,差异均有统计学意义(P〈0.05)。结论:宫颈癌组织中Bcl-x L、Bcl-2的高表达可能在宫颈癌抗凋亡机制中起重要作用,这对宫颈癌的易感性预测、早期诊断、疾病监测及其分子靶向治疗的开展具有重要指导意义。 Objective: To detect the expressions of apoptosis -related genes in cervical cancer among Uygnr women, explore the relationship between pathogenesis and clinicopathologieal parameters of cervical cancer. Methods: From June 2012 to March 2013, 21 nor- mal cervical specimens from the patients with uterine fibroids after biopsy and hysterectomy and 45 cervical cancer specimens in Department of Gynecology in Xinjiang Peopleg Hospital were collected ; real -time fluorescent quantitative PCR and immunohistochemical method were used to detect Bcl - xL and Bcl - 2 expression levels in cervical cancer and normal cervix; the relationship between expression levels of Bcl - xL and Bcl - 2 and cervical cancer was analyzed. Results: Bcl - xL expression in cervical cancer was up - regulated, which was corre- lated with diameter of tumor, pathological grades, clinical stages, ,depth of myometrial invasion, and lymph node metastasis. Bcl -xL mRNA expression level in cervical cancer was 4. 68 times more than that in normal cervix, the expression level of Bel -2 mRNA in cervical cancer was lower than that in normal cervix. There were statistically significant differences in the positive expression rates of Bcl - xL protein and Bcl - 2 protein between cervical cancer and normal cervix (X2 = 12. 357, P = 0. 000 ; X2 = 5. 820, P = 0. 015) . In cervical cancer group, there were statistically significant differences in the expression levels of Bcl - xL protein and Bcl - 2 protein among the patients with different clinical stages, diameters of tumor, depths of myometrial invasion, pathological grades, and lymph node metastasis or not (P 〈 0. 05 ) . Conclusion : High expressions of Bcl - xL and Bel - 2 in cervical cancer patients may play important roles in antiapoptotic mechanisms of cervical cancer, which have importmlt guiding significance for susceptibility prediction, early diagnosis, disease monitoring, and development of molecular targeted therapy of cervical cancer.
