摘要
【目的】筛选针对海岛棉、陆地棉与海岛棉之间的核心引物。【方法】以8个海岛棉和4个陆地棉品种(系)为材料,采用分子标记技术,对已在Cotton Microsatellite Database(CMD)网站上公布的3 229对SSR引物,PCR扩增,通过PAGE胶电泳、检测,对获得的扩增条带清晰、重复性好且分辨能力强的核心引物进行统计分析。【结果】94对为海岛棉核心引物,214对为陆海之间差异明显的核心引物。【结论】筛选到了部分核心引物,所选择的SSR引物在海岛棉中多态性较低,选出率仅为2.9%。
【Objective】The projects aims to screen the core primers between upland cotton and sea- island cotton.【Method】In this experiment,there were eight sea island cotton and four upland cotton taken as testing materials,and molecular markers was applied to test the 3 229 pairs SSR primers that had been published on the website Cotton Microsatellite Database( CMD) bv means of PCR amplification and PAGE electrophoresis in order to get clear amplified bands with reproducible and strong ability to distinguish core primers for statistical analysis.【Result】94 pairs belonged to sea island cotton core primers and 214 pairs belonged to core primers with significant differences between upland cotton and sea island cotton. 【Conclusion】Through the series of experiments,some core primers were screened out,but owing to the low cotton polymorphism of the selected SSR primers,the percentage of the filtration was only 2. 9.
