混合血样DNA等位基因分型探究

摘要 以人体混合血样为研究对象,研究两个体混合血样的基因座等位基因分型表现。实验材料与方法:采用Chelex-100法提取DNA,ID试剂盒进行PCR扩增,AB-3130基因自动分析仪电泳,Genemapper3.2软件进行分析。实验结果:两个体混合血样的基因座等位基因可表现为4个等位基因,3个等位基因,2个等位基因或1个等位基因。随着混合比例差距的增大,量少个体的等位基因峰相对于量多个体等位基因峰越来越低。当混合比例达到1:10时,量少个体的等位基因峰很低,基本上不影响量多个体的等位基因分型。 Target: Research on allelic classification perfor mance of gene locus in two mixed human blood. Materials and Methods: using Chelex-100 to extract DNA, amplifying PCR in ID kit, making analysis by AB- 3130 gene electrophoresis automatic analyzer and Genemapper 3.2 software. Results: 4 or 3 or 2 or 1 alleles can be showed in two mixed blood gene locus. With the increasing of mixture ratio, the allele peak with less amounts becomes lower and lower than those with more amounts. When mixing proportion reaches 1:10, allele peak is lowest, basically does not affect allelic classification in large amounts
