和厚朴酚抑制9L胶质瘤细胞增殖和诱导其凋亡的作用

Effects of Honokiol on inhibiting proliferation and inducing apoptosis of 9L gliosarcoma cells in vitro

目的探讨和厚朴酚(HNK)抑制9L胶质瘤细胞增殖并诱导其凋亡的作用。方法不同浓度HNK对9L胶质瘤细胞作用后,采用MTT实验评估9L细胞增殖水平,用细胞爬片Hoechst33258荧光染色及DNA Ladder检测细胞的凋亡情况,采用流式细胞术检测HNK对9L细胞周期的影响。结果 HNK呈剂量依赖性抑制9L细胞增殖,其48h的IC50为15μg/ml;HNK处理过的9L细胞,经Hoechst 33258染色后细胞核出现明显的凋亡形态学变化,并在DNA琼脂糖凝胶上可见特征性的细胞凋亡梯状条带;HNK阻滞9L细胞周期于G0/G1期。结论 HNK具有抑制9L胶质瘤细胞增殖并诱导其凋亡的作用。

Objective To assess the effects of Honokiol on inhibiting proliferation and inducing apoptosis of 9L gliosarcoma cells in vitro.Methods After the 9L cells cultured with different concentrations of Honokiol,the cell proliferation was assessed by MTT colorimetrie assay,cell apoptosis was determined by Hoechst 33258 fluorescent staining and DNA ladder,and cell cycle was examined by flow cytometry.Results The growth of 9Lcells was obviously inhibited by Honokiol in a dose-dependent manner with an IC50 of 15μg/ml at 48 hours.Typical morphologic changes of apoptosis were observed by Hoechst 33258 fluorescence staining and genomic DNA displayed a characteristic ladder pattern on agarose gel electrophoresis.Honokiol blocked the 9Lcell cycle in G0/G1 phase.Conclusion Honokiol can inhibit the proliferation and induce the apoptosis of 9L gliosarcoma cells in vitro.
