WNT信号通路研究进展 被引量:7

摘要 WNT信号通路参与胚胎发育、组织稳态及细胞癌变等。早期有研究显示,WNT信号作为脊椎动物发育早期一个关键的调节因子,调节胚胎干细胞分化必需的内胚层和中胚层谱系,维持成年动物和人体器官干细胞的自我分化等。经典WNT信号通路通过调节原癌基因β-catenin的稳定性,使β-catenin基因产物在多种癌细胞中异常活化。大规模测序工作发现了WNT信号通路中几个新的常见突变位点,是由WNT通路进行正性或负性调节,这就表明WNT信号在癌变过程中具有复杂的机制。本文叙述了以影响β-catenin为路径的相关信号转录因子的最新进展与成果,以加深学者对WNT信号通路的了解,为研发有效的新型抗肿瘤药奠定了坚实基础。
