不明原因不孕妇女种植窗期血小板活化因子及其受体的表达与子宫内膜超声类型的关系

Expression of platelet-activating factor and its receptor in women with unexplained infertility at implantation window stage and their relationship with ultrasonographic pattern of the endometrum

目的探讨不明原因不孕妇女种植窗期血小板活化因子(PAF)及其受体(PAF-R)的表达与子宫内膜超声类型的关系。方法选择不明原因不孕妇女30例,根据其自然周期尿促黄体生成素峰日子宫内膜类型分为C型子宫内膜10例(C型组)和A型子宫内膜20例(A型组)。在种植窗期,采用酶联免疫吸附试验测定法检测2组患者血清PAF水平;流式细胞术检测外周血单核细胞(PBMCs)PAF-R的表达;免疫组织化学法检测PAF-R在子宫内膜及内膜血管中的表达。结果不明原因不孕妇女C型组种植窗期血清PAF水平、PAF-R在PBMCs、子宫内膜和内膜血管中的表达较A型组均显著降低(P<0.05)。2组患者PBMCs中PAF-R的表达与子宫内膜组织中PAF-R的表达均呈正相关(P<0.05)。结论不明原因不孕妇女子宫内膜超声类型与种植窗期PAF及PAF-R的水平有关。PBMCs中PAF-R的表达水平可间接反映子宫内膜组织中PAF-R的表达情况。

Objective To investigate the expression of platelet-activating factor（ PAF） and its receptor（ PAF-R） in women with unexplained infertility at implantation window stage and their relationship with ultrasonographic pattern of the endometrium. Methods Thirty unexplained infertility women were divided into two groups according to their endometrium pattern on the peak day of urine luteinizing hormone of the natural cycle,including 10 cases of the endometrium pattern C（ pattern C group） and 20 cases of the endometrium pattern A（ pattern A group）. During the implantation window stage,enzyme linked immunosorbent assay was applied to analyze the level of PAF in serum. Expression of PAF-R in peripheral blood monocytes（ PBMCs） was examined by flow cytometry. PAF-R in endometrium and endometrial blood vessels was examined by immunohistochemistry methods. Results The level of PAF in serum,the expression of PAF-R in PBMCs,endometrium and endometrial blood vessels in the pattern C group were significantly lower than those in the pattern A group. PAF-R in PBMCs was positively correlated to that in the endometrium in the two groups. Conclusions The ultrasonographic pattern of the endometrium of unexplained infertility women is correlate to the concentration and expression of PAF and PAF-R at implantation window stage. The expression of PAF-R in PBMCs can indirectly reflect the expression of PAF-R in the endometrium.
