摘要
目的:检测黄连含药血清对骨髓源巨噬细胞增殖的影响,探讨相应血清药理学实验中含药血清的合适浓度。方法:采集小鼠骨髓细胞诱导其单核细胞分化为巨噬细胞,流式细胞仪检测CD11b、F4/80检测细胞纯度;采用黄连水煎剂灌胃干预清洁级SD大鼠,常规制备含药血清,MTT法检测黄连含药血清对巨噬细胞增殖的影响。结果:与同浓度的正常血清对照组OD值比较,黄连含药血清在10%~40%浓度范围内对细胞增值无明显影响(P〉0.05)。结论:在黄连含药血清干预巨噬细胞的相关实验中,含药血清加样浓度可选择在40%以内。
Objective: To analyze the effects of serum containing Huang-lian on proliferation of bone mar- row's macrophage, and to search for optimal concentration of serum medicine in pharmacology experiments. Method : Monocytes of bone marrow from mouse was extracted and induced to macrophages. The cell purity was determined by CD1 l b and F4/80 using flow cytometry. SD rats were stomach-fed with Huang-lian water decoc- tion. Their serum containing medicine was prepared with standard protocol. The effect on macrophage prolifera- tion was tested using MTT assays. Serum containing Huang-lian in 10% from 40% range had no significant effect on macrophage proliferation ( P 〉 0.05 ) , compared with OD value of control group incubated by normal serum. Conclusion: In experiments involving macrophages intervened by serum containing Huang-lian, the ser- um with concentration below 40% could be selected.
