摘要
目的研究番石榴叶总黄酮对MDA-MB231肿瘤细胞体外抑制及细胞凋亡的诱导作用。方法以番石榴叶总黄酮为原料,采用体外细胞培养方法,MTT法检测其对MDA-MB231肿瘤细胞株的生长抑制率,流式细胞仪检测总黄酮作用下肿瘤细胞的凋亡率。结果番石榴叶总黄酮对MDA-MB231肿瘤细胞株的生长具有较强的抑制作用。总黄酮对肿瘤细胞的IC50为270μg/ml,根据番石榴总黄酮的不同给药剂量逐渐变化,测得的细胞凋亡率也呈现逐步变化过程,数值分别为2.0%、15.7%、22.1%、26.2%。结论番石榴叶总黄酮醇在体外可显著抑制MDA-MB231肿瘤细胞株的生长,有效抑制细胞的增殖作用,可能与其对细胞的凋亡机制的调控有关。
Objective To explore the inducible effect of Guava leaves flavonoids on the antitumor effect in v itro of MDA-MB231 and cell apoptosis. Methods The MTT methods was used to detect the cell growth inhibition rate,the cell apoptosis of MDA-MB231 was measured by the flow cytometry. Results The proliferation of MDA-MB231 cells was obviously restrained by the use of total Flavones in concentration and time dependent characteristics in v itro,the value of the inhibition concentration (IC50) was 270 μg/ml,the apoptosis-rate was 2.0%,15.7%,22.1%,26.2% respectively. Conclusion The use of total flavones of Psidium guajava leaves can significantly inhibit the growth of MDA-MB231 cells,perhaps,the action is relevant to the changes of the cell apoptosis way.
