不同方式运动对生长期雄性小鼠骨形成和骨吸收代谢影响的研究 被引量:8

Effects of Different Kind of Exercises on the Metabolism of Bone Formation and Bone Restoration in Growth of Male Mice

摘要 目的探讨不同方式运动对生长期雄性小鼠骨形成和骨吸收代谢的影响及其可能机制。方法 24只4周龄雄性C57BL/6小鼠随机分为对照组(C组)、游泳组(S组)和下坡跑组(D组)。8周训练结束后,取右侧下肢骨并利用RT-PCR技术检测骨中Runx2、Osx、NFATc1和Cath K的mRNA表达;取血和左侧下肢胫骨并利用比色法和骨组织切片染色检测血清和骨中ALP和TRAP活性;取左侧前肢骨并利用Hologic Discovery A骨密度仪检测肱骨远侧端骨密度。结果与C组相比,S组血清(P<0.01)和骨中ALP活性升高、NFATc1(P<0.05)和Cath K(P<0.05)的mRNA表达下调;与C组相比,D组血清(P<0.01)和骨中ALP活性均升高,Runx2(P<0.01)和Osx(P<0.05)的mRNA表达上调,而血清(P<0.05)和骨中TRAP活性均下降,NFATc1(P<0.01)和Cath K的mRNA(P<0.01)表达下调,BMD(P<0.01)升高;与S组相比,D组血清(P<0.01)和骨中ALP活性均升高,Runx2(P<0.05)和Osx(P<0.05)的mRNA表达上调,而血清(P<0.05)和骨中TRAP活性均下降,NFATc1mRNA(P<0.05)和Cath K mRNA(P<0.05)表达下调,BMD(P<0.01)升高。结论下坡跑运动可通过促进骨形成并抑制骨吸收,从而促进生长期小鼠的骨生长发育,且其作用效果优于游泳。 Objective Research the effect and mechanism of different kind of exercises on bone formation and bone restoration.Methods 24 4-week old male C57 BL /6 mice were randomly divided into three groups,control group(C group),swimming group(s group) and downhill running group(D group).After 8-week training,Use the method of RT-PCR to test the mRNA expression of Runx2,Osx,NFATc1 and Cath K in the right femur.Use the method of ELLISA and tissue slice staining to test the enzymatic activity of ALP and TRAP in the mice serum and bone.Use the Hologic Discovery A to detect the left humerus BMD of forelimb.Results Compared to C group,ALP enzymatic activity in serum(P〈0.01) and bone was significantly increased and the mRNA expression of NFATc1(P〈0.05) and Cath K(P〈0.05) were significantly downregulated in S group.Compared to C group,the Runx2mRNA(P〈0.01),Osxm RNA(P〈0.05) and ALP enzymatic activity in serum(P〈0.01) and bone were significantly up-regulated,the NFATc1mRNA(P〈0.01),Cath K mRNA(P〈0.01) and TRAP activity in serum(P〈0.05) and bone were all significantly decreased,bone mineral density(P〈0.01)was significantly increased in D group.Compared to S group,the mRNA expression of Runx2(P〈0.05) and Osx(P〈0.05) and ALP activity in blood(P〈0.01) and bone were all significantly increased,the mRNA expression of NFATc1(P〈0.05) and Cath K(P〈0.05) were both significantly down-regulated,the bone mineral density(P〈0.01) was significantly increased in D group.Conclusion Exercise increased the bone growth and development by promoting bone formation and inhibiting bone restoration,but the downhill running had better effect on bone metabolism than swimming.
