介入栓塞法小型猪急性心肌梗死模型的建立 被引量:4

Establishment of acute myocardial infarction model by interventional embolization in mini-pigs

摘要 目的:用介入栓塞法建立小型猪的急性心肌梗死模型。方法:11头贵州种小型猪,体重30~45 kg。术前采集血液进行实验室检查(心肌酶谱和肌钙蛋白等);并行心电图及超声心动图检查。常规冠状动脉造影后,经导管将栓子推送到左冠状动脉前降支或回旋支远端进行栓塞,复查造影结果满意后结束手术操作,并复查心电图。术后24 h采集血液复查实验室指标,并复查超声心动图。术后5周处死实验动物行心脏病理组织学检查。手术前后的实验室检查指标及心功能指标(射血分数和短轴缩短率)采用配对t检验。结果:11头小型猪中,有8头成功建立急性心肌梗死模型,建模成功率72%。栓塞前:各项检查结果正常。栓塞后:冠状动脉造影检查栓塞远端未见显影;心电图出现不同程度的异常改变;实验室检查及心功能检查与术前比较,其差异具有统计学意义(P〈0.05);术后病理证实均有梗塞灶的存在。结论:介入栓塞法成功率高,简单易行,是建立稳定心肌梗死模型的较好方式之一。 Objective: To investigate the level of protein kinase Cα( PKCa),oncofetal fibronectin( oncofetal FN) in human mesangial cells( HMCs) exposed to high glucose,and explore the relationships of PKCa with expression of oncofetal FN mRNA. Methods:HMCs were divided into the following groups: normal glucose group( NG,D-glucose 5 mmol / L),osmotic control group( LG,D-glucose 5mmol / L + L-glucose 20 mmol / L),high glucose group( HG,D-glucose 25 mmol / L),and high glucose + Safingol group( HS,D-glucose25 mmol / L + Safingol 40 μmol / L). The expression and translocation of PKCa protein was observed by confocal laser scanning microscopy,The mRNA expression of oncofetal FN were detected by RT-PCR. Results:( 1) High glucose induced PKCa translocation from cytosol to nucleus,and quantitative analysis showed the ratio of plasma / nuclear fluorescence intensity in HG group decreased 20% compared with NG group( P〈0. 05). Oncofetal FN mRNA in HG group was up-regulated,with 2. 36 folds of NG group( P〈0. 05).( 2)Compared with HG group,the activation of PKCa in group HS was inhibited and the ratio of plasma / nuclear fluorescence intensity increased 15%. Besides,the mRNA expression of oncofetal FN in group HS was 45% down-regulated compared with group HG( both P〈0. 05). Conclusion: High glucose increase the expression of oncofetal FN in HMCs,which related to the activation of PKCa.
