摘要
目的 探讨信号传导与转录激活因子3和血管内皮生长因子在皮肤恶性黑色素瘤中的表达及临床意义.方法 采用组织芯片免疫组化学S-P法、免疫印迹法检测信号传导与转录激活因子3及血管内皮生长因子在35例皮肤恶性黑色素瘤、30例色素痣组织中的表达情况.结果 在皮肤恶性黑色素瘤组织中,信号传导与转录激活因子3、血管内皮生长因子的表达阳性率分别为65.7%、60.0%,均明显高于在色素痣中表达的阳性率(P<0.01).皮肤恶性黑色素瘤中信号传导与转录激活因子3和血管内皮生长因子表达均与肿瘤的细胞生长、浸润深度和淋巴结转移呈正相关(P<0.05);信号传导与转录激活因子3和血管内皮生长因子的表达呈显著正相关(P<0.001).结论 信号传导与转录激活因子3可能通过上调血管内皮生长因子的表达,促进皮肤恶性黑色素瘤组织中新生血管的形成,与肿瘤生长、侵袭和转移呈密切相关.为皮肤恶性黑色素瘤的靶向性治疗提供新思路.
Objective To explore the expression of signal transducers-transcription activators 3 and vascular endothelial growth factor in cutaneous malignant melanoma and its clinical significance. Methods The expression of signal transducers-transcription activators 3 and vascular endothelial growth factor were examined in 35 cases of cuta- neous malignant melanoma and 30 cases of nevus by immunohistochemistry and Western blot. Results The positive rates of signal transducers-transcription activators 3 and were 65.7% and 60.0% in the cutaneous malignant melano- ma, which were significantly higher than those which were 15.4% and 3.3% in the nevus ( P〈0.01 ). Both the ex- pression of signal transducers-transcription activators 3 and vascular endothelial growth factor were significantly corre- lated to growth of tumor cell, depth of tumor invasion and tumor lymph node metastasis ( P 〈 0.05 ). Moreover, a significant con'elation was found between the expression of signal transducers-transcription activators 3 and vascular endothelial growth factor in cutaneous malignant melanoma (P〈0.001). Conclusion The signal transducers-tran- scription activators 3 may promote the formation of new blood vessels in the CMM by up-regnlating the expression of vascular endothelial growth factor which was closely correlated to the tumor growth, invasion and metastasis. This study will provide new way for the targeted treatment of cutaneous malignant melanoma.
