摘要
目的对小凹蛋白1(caveolin 1,Cav-1)在不同肿瘤中的表达差异及作用机制的研究进展进行综述分析。方法应用PubMed及CNKI期刊全文数据库检索系统,以"小凹蛋白和癌"等为关键词,检索1995-01-2013-12的相关文献,共检索到英文文献585条,中文文献30条;纳入标准:1)胃癌、肝细胞癌、子宫颈癌、乳腺癌、前列腺癌及白血病;2)Cav-1介导的信号通路;3)Cav-1介导的内吞作用;4)Cav-1与自噬。根据纳入标准符合分析文献21篇。结果小凹蛋白在不同的肿瘤及肿瘤的不同阶段中表现出差异性表达,即在胃癌组织、子宫颈癌和乳腺癌组织中表现为低表达,而在肝细胞癌、前列腺癌和白血病组织中表现为高表达。其作为细胞信号转导的调控中枢,影响着肿瘤细胞的生物学活性,它的突变、缺失与肿瘤的发生、浸润及转移密切相关。结论 Cav-1与肿瘤发生发展的关系紧密,并且其调控作用贯穿于机体正常组织发生癌变的各个环节,对小凹蛋白的不断研究以及配合大量临床资料的分析将会为肿瘤的早期诊断、治疗以及预后判断提供一条新的策略。
OBJECTIVE To study the differential expression and mechanisms of caveolin 1 (Car-l) in different tumors. METHODS PubMed and CNKI database have been searched with Caveolin and cancer as keywords,and a total of S8g English references,30 Chinese references from 1998-01 to 2013-12. Inclusion criteria: 1) gastric cancer,hepatocellular carcinoma,carcinoma of uterine cervix,breast cancer,prostate cancer and leukemia;2) Cav-l-mediated signaling pathway; 3) Cav-l-mediats endocytosis;4) Cav-1 and autophagy. 21 references met the inclusion criteria. RESULTS Caveolin per- forms differential expressions at different stages of different tumors. The expression level of Cav-1 in gastric cancer,cervi- cal cancer and breast cancer is relative low,and the expression level of Cav-1 in hepatocellular carcinoma, prostate cancer and leukemia is relative high. As the key factor of cellular signal transduction regulatory network, affecting the biological activity of tumor cells, and its mutations, deletions are deeply effected tumorigenesis, invasion and metastasis of the tumors. CONCLUSIONS Caveolin and tumorigenesis has close relationship,and it runs through all aspects of the regula- tion of the bod/s normal tissue cancer. Continuous research on caveolin with a large number of clinical data will provide a new strategy for the early diagnosis of the tumor,treatment and prognosis.
