摘要
目的:研究川乌在加热前后生物碱变化。方法:采用LC-MS测定方法,Agilent Zorbax Extend-C18色谱柱(250 mm×4.6 mm,5μm);柱温为30℃;流动相为醋酸铵(氨水调pH 8)-乙腈(20 mmol·L-1);流速为1.0 mL·min-1;质谱采用正离子检测模式。结果:分析鉴定了川乌中13个生物碱类成分,其中9个生物碱在加热过程中的变化较明显。结论:本方法简便,具有较高的准确度和精密度,可用于生物碱变化的分析。
Objective: To analyze the changes of chemical constituents in Radix Aconiti Preparata before and after heating. Methods: The different extract of Radix Aconiti Preparata were detected by using HPLC/ESI-MS. Results: The constituents with significant differences before and after heating were identified. The results showed that a total of 13 chemical constituents were identified and nine constituents were significantly different before and after heating. Conclusion: The HPLC/ ESI-MS method is proved to have a high sensitivity and the result is accurate and reliable. The method could reflect the changes of multiple chemical constituents of Chinese materia medica in the course of processing.
