摘要
目的 :评价拓扑替康对人新鲜乳腺癌细胞的抗癌效果 ,并将其与 5种乳腺癌常用化疗药物对人新鲜乳腺癌细胞的抑制作用进行对比。方法 :以 2 0例已确诊的新鲜乳腺癌癌块为实验对象进行人新鲜乳腺癌细胞培养药物敏感性试验。结果 :(1)人乳腺癌细胞原代培养结果 :2 0例乳腺癌标本中 ,阳性率 85 % (17/ 2 0 ) ,获得 1× 10 5/ m L的乳腺癌单细胞悬液 10 m L以上。 (2 )原代培养人乳腺癌细胞对 6种抗癌药物敏感性试验结果 :常规浓度组敏感率依次为 :PTX>ADM>TPT>MTX>5 - Fu>DDP。 (3)统计学分析χ2 检验 ,TPT高、中浓度组之间差异无显著性 (P>0 .0 5 ) ,两者与低浓度组之间差异有显著性 (P<0 .0 5 )。结论 :(1)拓扑替康对乳腺癌有效 ,相当于氨甲蝶呤和 5 -氟尿嘧啶的作用。 (2 )乳腺癌细胞培养药物敏感性试验作为临床筛选新的抗乳腺癌药物的方法之一 ,敏感性较高 。
Objective:To test the antitumor activity of topotecan to human fresh breast cancer cell and to compare the inhibitive effects of topotecan with the other five effective antitumor agents to breast cancer.Methods:20 breast cancer specimens were regarded as the test object to incubate in primary culture.Results:1.Primary culture result of human fresh breast cancer cell:In 20 specimens, culture positive rate was 85 %(17/20).2.The in vitro chemosensitivity of breast cancer cell to 6 antitumor agents: In normal concentration:PTX>ADM>TPT>MTX>5 Fu>DDP.3.Statistical analysis:χ 2 test demonstrated that topotecan had no significant variant between the higher and normal concentration groups( P >0.05),but they had significant variant with the lower concentration.Conclusion:1.Topotecan is a new effective antitumor agent against breast cancer.Its chemosensitivity is equal to methotrexate and fluorouracil. 2.The drug screening assay of fresh human breast cancer should be spreaded further.
