摘要
为确定云南林科院普文林场葡萄柚试验园从美国佛罗里达州引种的葡萄柚品种是否感染黄龙病以及其病原类型,采集116个柑桔黄龙病疑似病样进行PCR检测、16SrDNA序列测定及系统进化分析。结果表明,116个样品中共有96个样品感染黄龙病,总感病率达到82.8%;在该试验园分离出的黄龙病菌16SrDNA的序列一致性达到100%,与NCBI中的柑桔黄龙病亚洲种(Candidatus Liberibacter asiaticus)、柑桔黄龙病非洲种(Candidatus Liberibacter africanus)和柑桔黄龙病美洲种(Candidatus Liberibacter americanus)序列的相似性分别为100%、97.6%-98.5%和96.0%-96.7%。该试验园葡萄柚上的柑桔黄龙病菌属于亚洲种。
In order to determine whether the grapefruit varieties introduced from Florida have been infected with Huanglongbing and the types of pathogen in Puwen grapefruit experimental orchard of Forest Science Academy of Yunnan province,116 citrus samples suspected to have huanglongbing were tested by PCR,16 SrDNA sequence analysis.The results showed that 96 samples were infected with huanglongbing pathogen with infection ratio at 82.8%.The isolated pathogen 16 SrDNA showed 100% sequence similarity to each other,100% to Candidatus Liberibacter asiaticus,97.6% to 98.5% to Candidatus Liberibacter africanus,and 96.0%to 96.7% to Candidatus Liberibacter americanus,respectively.The results indicated that the HLB pathogen in grapefruit experiment orchard of Puwen is Ca.L.asiaticum.
