摘要
利用测序方法测定了橄榄品种(系)的核糖体DNA中的内转录间隔区(ITS)序列(包括ITS1、5.8S和ITS2)。结果表明,橄榄ITS区序列总长度629-634bp,长度变异仅为5bp。其中ITS1区为232-237bp,ITS2区为234-239bp,5.8SDNA均为165bp,且高度保守无变异位点。橄榄品种(系)间ITS序列变异位点较少,仅有17个变异位点,占总碱基数的5.93%,简约信息位点7个,占总碱基数1.51%,单一信息位点10个,占总碱基数的2.16%,并且ITS1序列较ITS2序列变异丰富。因此,利用ITS序列中变异位点可以作为特异DNA指纹鉴定位点,为部分品种(系)的鉴定依据,但是仅仅利用ITS序列还难以鉴定区别所有橄榄品种资源。
The sequences of ITS(including ITS1,5.8Sand ITS2)of ribosomal DNA of Canarium album L.were determined by sequencing method.The results showed that the total length of ITS varied from 629 to 634bp(ITS1from 232 to 237bp,ITS2 from 234to 239bp),and the variation of length was only 5bp.The length of 5.8SDNA was 165 bp without the variation.Among different varieties of olive,there were only seventeen variable loci in ITS sequences,which were 5.93% of the entire sequence,7parsimony informative loci,which were 1.51%,and 10 singleton variable loci,which were 2.16%.The variances of ITS1 sequence were richer than those of ITS2.Therefore,ITS variable loci can be used as DNA molecular marker for the identification of some cultivars,but still not able to identify all Canarium album L.cultivars resources only based on ITS sequence.
