摘要 目的:建立同时测定静脉给药山楂叶提取物后大鼠血浆中三种黄酮含量的HPLC方法。方法:采用HPLC,以黄芩苷为内标物,甲醇沉淀蛋白,Phenomsil C18(5μm,250×4.6 mm);流动相:甲醇-乙腈-四氢呋喃-0.4%醋酸水(6∶1.5∶18.5∶74);检测波长:332 nm。结果:实验表明VG药动参数AUC0→t与剂量的增长呈正比,VR、VIT在大鼠血浆中的AUC0→t与给药剂量不成正比,表明VR、VIT在大鼠血浆中遵循非线性药代动力学行为。结论:建立HPLC测定大鼠体内3种黄酮方法简单、专属性强,适用于黄酮类成分大鼠体内的药代动力学研究。 Objective: To establish the HPLC method for simultaneous determination of three kinds of flavonoids contents in rats plasma after intravenous administration of hawthorn leaf extract. Methods: The plasma samples were deproteinized with methanol after addition of internal standard(I. S.) baicalin. HPLC analysis was performed on a Phenomsil C18(5μm,250 mm × 4. 6mm) analytical column,using methanol-acetonitrile-tetrahydrofuran-0. 4% aqueous acetic acid(6: 1. 5: 18. 5: 74) as the mobile phase with UV detection at 332 nm. Results: The experiment results showed that pharmacokinetic parameters AUC0→tincreased with the VG doses. The AUC0→tof VR and VIT in rats plasma were not directly proportional to the administration dose.The results indicated that the pharmacokinetics of VR and VIT in rats obeyed nonlinear processes. Conclusion: The simple and specific HPLC method can be successfully applied during a pharmacokinetic study of flavonoids in rats plasma.
