生莱菔子和炒莱菔子中芥子碱硫氰酸盐的含量测定 被引量:4

摘要 目的建立用HPLC法测定生莱菔子和炒制莱菔子中芥子碱硫氰酸盐含量的方法,比较不同产地药材中芥子碱硫氰酸盐的含量,并研究炒制对药材中芥子碱硫氰酸盐含量的影响。方法以苯基硅烷键合硅胶为填充剂;进样量5μL;以乙腈-3%冰醋酸溶液(15∶85)为流动相;检测波长326 nm。结果芥子碱硫氰酸盐进样量在0.047 2~1.888 0μg之间有良好线性关系;生莱菔子的加样平均回收率为99.30%,RSD=0.4%(n=6);炒制莱菔子的加样平均回收率为94.58%,RSD=0.3%(n=6)。结论本法快速、简便,结果准确、可靠;不同产地药材芥子碱硫氰酸盐含量差异大;炒制后莱菔子中芥子碱硫氰酸盐含量有所增加。 Objective To establish a method for content determination of raw and frying semen raphani,compare the content of samples from different regions of sinapine thiocyanate,and explore the effect of frying on sinapine thiocyanate.Methods Phenyl silane bonded silica column was used.The flow rate was 5 μL.The mobile phase was acetonitrile-3%acetate(15:85).The detection wavelength was set at 326 nm.Results The linear range of sinapine thiocyanate was 0.047 2~1.888 0 μg.The average recovery of raw semen raphani was 99.30%,RSD=0.4%(n=6),and the average recovery of frying semen raphani was 94.58%,RSD=0.3%(n=6).Conclusion The method is rapid,simple,accurate,and reproducible.It has marked discrepancy in the content of samples from different regions.Frying Semen Raphani can increase the content of Sinapine thiocyanate.
