摘要
目的 探讨调节性T细胞(Treg)与实验性自身免疫性肌炎(EAM)的关系.方法 将20只BALC/c小鼠完全随机分为EAM组和刘照组,各10只.EAM组皮下注射0.25 ml弗氏完全佐剂+0.25 ml20 μg/μl骨骼肌匀浆,对照组皮下注射0.25 ml弗氏完全佐剂+0.25 ml磷酸盐缓冲液,观察2组小鼠肌肉组织形态学变化和CD45表达.流式细胞仪检测2组小鼠外周血及脾脏单个核细胞中CD4+CD25+ Foxp3+ Treg所占CD4+T细胞的比例.结果 EAM组小鼠肌肉组织HE染色可见大量炎性细胞浸润,伴有肌纤维萎缩、坏死、吞噬,CD45表达阳性.对照组未见炎性细胞浸润.EAM小鼠外周血单个核细胞中CD4+CD25+Foxp3Treg所占CD4 T细胞的比例低于对照组小鼠[(8.0±4.O)%比(15.5±6.9)%],脾脏单个核细胞中CD4+CD25 Foxp3+ Treg所占CD4 T细胞的比例低于对照组小鼠[(4.7±2.2)%比(8.6±4.0)%],差异均有统计学意义(均P<0.05).结论 Treg细胞群在维持机体免疫平衡中起重要作用,为特发性炎性肌病免疫机制的研究及治疗提供了新思路.
Objective To observe the proportion of CD4 CD2~ Foxp3 regular T cells (Tregs) for CD4 T ceils in peripheral blood and spleen mononuelear ceils in a mouse model of experimental autoimmune myositis (EAM) ; to explore the relation between Tregs and autoimmune myositis, Methods Totally 20 BALC/c mice were randomly divided into EAM group administrated with Freund's complete adjuvant ( CFA, 0.25 ml) + skeletal muscle homogenates (20 μg/μl, 0.25 ml), and control group that had CFA (0.25 m]) + phosphate buffer solution (0.25 ml). The morphological changes of muscle tissue and expression of CD45 were observed, and the proportions of CD4+ CD2+ Foxp3 Tregs for CD4 T ceils in peripheral blood and spleen mononuelear cells were measured by flow eytometry. Results In EAM group, HE staining showed abundant inflammatory cell infiltration with expression positive of CD45 , accompanied by atrophy, necrosis, phagocytosis of muscle fiber; no inflammatory cell infiltration was observed in control group. The proportions of CD4 CD2+ Foxpf Tregs for CD4 T cells in peripheral blood and spleen mononuelear cells in EAM group were both significantly lower than those in control group[ (8.0± 4.0)% vs(15.5±6.9)%, (4.7±2.2)% vs (8.6±4.0)%,P〈0.05]. Conclusion Treg cells play an important role in maintaining immune balance, which provides a new idea for research and treatment of the idiopathic inflammatory myopathies.
