摘要
研究已经证实水稻线粒体基因组异常嵌合基因orf H79和orf79分别为HL-CMS和BT-CMS基因。为探讨orf H79在水稻资源中的遗传及变异,对来自不同稻作国家的124份水稻材料进行orf H79特异性PCR检测。有18份水稻材料检测出含有HL-CMS基因orf H79及其同源序列,结果表明HL-CMS基因orf H79及其同源序列在稻种资源中具较高频率的分布(14.5%)。DNA序列分析显示HL-CMS基因orf H79及其同源序列和BTCMS基因orf79具有非常保守的遗传特性(97%),共检测出6个多态性碱基位点,6个位点碱基的变异导致多肽ORFH79产生了5个位点氨基酸的变化。基于orf H79DNA序列的聚类分析显示18份含orf H79的水稻材料被分成了4个类群。多肽ORFH79的氨基酸序列变异分析表明HL-CMS胞质在稻种资源中产生了明显的遗传分化,至少存在3种以上的变异类型。研究结果为新型水稻CMS胞质的发掘与培育提供了理论和实验依据。
The aberrant chimeric gene in rice mitochondrial genome,orf H79 and orf79,was proved to be the gene of HL-CMS and BT-CMS,respectively.In order to survey the heredity and variation of orf H79 in rice resources,124 rice varieties( accessions) from different countries were detected with PCR amplification.18 rice varieties( accessions) were found to share the gene orf H79 of HL-CMS and its homologous sequences. The results indicated that the orf H79 and its homologous sequences had a relatively high-frequency distribution in rice resources( 14. 5%). The analysis of DNA sequences showed that the orf H79 / orf79 and its homologous sequences had a very conservative hereditary trait in rice resources( 97%).6 polymorphism variation sites of base were detected in DNA sequences of orf H79 and its homologous sequences,which led to five variation sites of amino acid in polypeptide ORFH79.The clustering analysis based on DNA sequences of orf H79 and its homologous sequences showed that the 18 rice varieties( accessions) fell into four groups.The variation analysis of amino acid sequences indicated that the rice HL-CMS cytoplasm had distinct differentiation and at least more than three variation types in rice resources.The results will provide important theoretical and experimental basis for excavation and cultivation of new rice CMS lines.
